Purpose Many in vitro studies of the analysis of the lactoferrin (LF) effect on cells have been reported. However, no study has yet investigated the effect of LF on osteogenic differentiation of human adipose-derived stem cells (hADSCs). The aim of this study was to evaluate the effect of LF on osteogenic differentiation of human adipose stem cells.Methods The hADSCs were cultured in an osteogenic medium with 0, 10, 50 and 100 μg/ml LF, respectively. hADSC proliferation was analysed by Cell Counting Kit-8 (CCK-8) assay, and cell osteogenic differentiation was evaluated by alkaline phosphatase (ALP) activity assay, von Kossa staining and real-time polymerase chain reaction (RT-PCR). Results Cell proliferation was significantly increased by LF in a dose-dependent manner from days 4 to 14. Cells cultured with 100 μg/ml LF presented a higher activity compared with the control. The deposition of calcium was increased after the addition of LF. The mRNA expression of type I collagen (COL-I), ALP, osteocalcin (OCN) and RUNX2 increased markedly as a result of LF treatment. Conclusions We have shown for the first time that LF could promote the proliferation and osteogenic differentiation of hADSCs, which could be a promising approach for enhancing osteogenic capacity of cell-based construction in bone tissue engineering.

Osteoblastic differentiation of stromal ST-2 cells from octacalcium phosphate exposure via p38 signaling pathway

The present study investigated the role of mitogen-activated protein kinase (MAPK) signaling in osteoblastic differentiation of stromal ST-2 cells induced by synthetic octacalcium phosphate (OCP) incubation. Since our previous studies revealed that OCP consumes calcium ions in media during conversion to hydroxyapatite, the effect of the ions on ST-2 cell differentiation with or without OCP crystals was analyzed. The effect of presence or absence of MAPK inhibitors was also analyzed. OCP increased alkaline phosphatase (ALP) activity and the mRNA expression of differentiation markers via the p38 signaling pathway. The PD98059 MAPK inhibitor increased ALP activity and differentiation marker genes in cells cultured in OCP-coated wells. Reduction of calcium ions in the medium by EGTA increased the ALP activity without OCP in the presence of phosphate ion concentrations up to 7.5 mM. OCP may enhance osteoblastic differentiation through the p38 signaling pathway via the reduction of calcium ions induced by its physicochemical property.

RESUMOObjetivo: Comparar a percentagem de neoformação óssea promovida pelo enxerto ósseo autógeno e três tipos de materiais de substituição de características distintas em cavidades em fêmures de ratos. Métodos: Foram realizadas duas cavidades de 5,4 x 2,7mm, em cada fêmur (direito e esquerdo), de 14 ratos Wistar isogênicos. Cada um dos quatro defeitos criados foi preenchido com o osso autógeno ou com um dos três materiais testados -hidroxiapatita (HA), Genphos ® (HA+ ß-TCP) e GenMix ® (um enxerto ósseo bovino composto). Ao final dos períodos de seis semanas (n = 6) e 12 semanas (n = 8), os animais foram sacrificados. As lâminas (coradas com Picro-Sirius) foram analisadas por microscopia óptica normal e software específico. Resultados: Os grupos com o osso autógeno se mostraram muito superiores aos demais nos dois tempos analisados, tendo média de neoformação óssea ± DP de 90,6 ± 10,8% em seis semanas, e 98 ± 9,2% em 12 semanas (p > 0,0001 em ambos os tempos analisados). Em seis semanas, os resultados para os demais grupos foram os seguintes: Genphos ® , 46 ± 7,1%; HA, 43,1 ± 8,4%; e GenMix ® , 57,3 ± 4,5%. Em 12 semanas: Genphos ® , 47,8 ± 11,1%; HA, 39,9 ± 5,4%; GenMix ® , 59,7 ± 4,8%, significativa (p = 0,007).
