Comparison of HCl extraction versos total iron analysis for iron tissue analysis

Abstract: Extraction of Fe from fresh leaves with 0.1 N HC1 proved to be a better indicator of the Fe status of a variety of ornamental tropical foliage and flowering plants compared to total Fe, 0.1 N HCl-ether, and IN HC1 extraction. It consistently gave higher correlations (r = 0.73 to 0.95 depending on the species) with chlorophyll concentration than the other methods tested. However, even 0.1 N HC1 extraction did not distinguish levels of Fe deficiency accurately when compared between species.

“…As the stability constant of Fe 2+ -phenanthroline complex is considerably greater than that of Fe 3+ -phenanthroline complex, Katyal and Sharma (1980) considered phenanthroline as Fe 2+ specific extractant. The finding of the present study therefore support the hypothesis that the fraction of Fe associated with chlorophyll synthesis is ferrous Fe (Katyal and Sharma, 1980;Lang and Reed, 1987).…”

“…According to Marschner (2003), a fraction of Fe that undergoes reversible Iron Deficiency in Avocado 39 ferrous/ferric Fe oxidoreduction is of particular physiological importance. However, several other authors suggested that the fraction of Fe associated with chlorophyll synthesis is ferrous Fe (Katyal and Sharma, 1980;Lang and Reed, 1987).…”

“…Total iron concentration has been found a good index of the iron nutritional status in experimental assays where conditions were similar for all treatments (Lang and Reed, 1987), although the importance of other indexes, such as the ratios between some nutrients, has been frequently mentioned in the literature. The P/Fe ratio in leaf as an iron status index was proposed by DeKock, 1981.…”

“…This fraction has been suggested to comprise Fe in the ferrous form (24). A number of methods using different extractants, chiefly weak acids or chelating agents, are now available for the determination of different fractions of Fe in plants (1,3,9,11,12,14,18,19,24).…”
