Comparison of Gene Expression of Umbilical Cord Vein and Bone Marrow–Derived Mesenchymal Stem Cells

Abstract: Mesenchymal stem cells (MSCs) give origin to the marrow stromal environment that supports hematopoiesis. These cells present a wide range of differentiation potentials and a complex relationship with hematopoietic stem cells (HSCs) and endothelial cells. In addition to bone marrow (BM), MSCs can be obtained from other sites in the adult or the fetus. We isolate MSCs from the umbilical cord (UC) veins that are morphologically and immunophenotpically similar to MSCs obtained from the BM. In culture, these cells … Show more

“…Previous comparative studies on proliferation rate, gene expression profile, differentiation potential, and efficiency of UCBMSC, fetal BMMSC, adult BMMSC, and adiposederived hMSCs have produced conflicting results, although all hMSC types have been demonstrated to show systematically similar cell surface antigen expression [16][17][18][19][20][21]. In this study, we focused on comparison of UCBMSCs and BMMSCs from different age groups.…”

“…Several comparative studies on differentiation potency, proliferation rate, immunophenotype, and gene expression of hMSCs from different sources have been previously conducted [16][17][18][19][20][21]. In general, cell surface antigen characteristics are similar for hMSCs derived from umbilical cord, fetal bone marrow, adult bone marrow, and adipose tissue; but inconsistencies in differentiation potential and proliferation rate have been reported.…”

“…1D). Since hematopoietic and mesenchymal stem cells derived from umbilical cord blood vs. bone marrow exhibit different gene expression profiles [34][35][36], these conflicting results may simply be due to the different model systems utilized. Despite these inconsistencies, the previous and current results suggest that expression of both CKLiK and CaMKKα are required throughout normal neutrophil development, but that attenuated expression may be important during late stages of neutrophil maturation.…”

“…Consistent with our results, MMP-2 mRNA expression has been previously detected in hMSCs isolated from bone marrow and umbilical cord blood. 32,48,49 Others had shown a cytoplasmatic localization of MMP-2 in hMSCs and its regulation by cancer-associated genes. 50 Although the investigators suspected the enzyme to be implicated in hMSC migration, they did not provide direct evidence for this assumption.…”
