Seventy authentic honey samples of 9 different floral types (rhododendron, chestnut, honeydew, Anzer (thymus spp.), eucalyptus, gossypium, citrus, sunflower, and multifloral) from 15 different geographical regions of Turkey were analyzed for their chemical composition and for indicators of botanical and geographical origin. The profiles of free amino acids, oligosaccharides, and volatile components together with water activity were determined to characterize chemical composition. The microscopic analysis of honey sediment (mellissopalynology) was carried out to identify and count the pollen to provide qualitative indicators to confirm botanical origin. Statistical analysis was undertaken using a bespoke toolbox for Matlab called Metabolab. Discriminant analysis was undertaken using partial least-squares (PLS) regression followed by linear discriminant analysis (LDA). Four data models were constructed and validated. Model 1 used 51 variables to predict the floral origin of the honey samples. This model was also used to identify the top 5 variable important of projection (VIP) scores, selecting those variables that most significantly affected the PLS-LDA calculation. These data related to the phthalic acid, 2-methylheptanoic acid, raffinose, maltose, and sucrose. Data from these compounds were remodeled using PLS-LDA. Model 2 used only the volatiles data, model 3 the sugars data, and model 4 the amino acids data. The combined data set allowed the floral origin of Turkish honey to be accurately predicted and thus provides a useful tool for authentication purposes. However, using variable selection techniques a smaller subset of analytes have been identified that have the capability of classifying Turkish honey according to floral type with a similar level of accuracy.

This work presents a RP-HPLC method for the simultaneous quantification of free amino acids and biogenic amines in liquid food matrices and the results of the application to honey and wine samples obtained from different production processes and geographic origins. The developed methodology is based on a pre-column derivatization with o-phthaldialdehyde carried out in the sample injection loop. The compounds were separated in a Nova-Pack RP-C 18 column (150 mm × 3.9 mm, 4 m) at 35• C. The mobile phase used was a mixture of phase A: 10 mM sodium phosphate buffer (pH 7.3), methanol and tetrahydrofuran (91:8:1); and phase B: methanol and phosphate buffer (80:20), with a flow rate of 1.0 ml/min. Fluorescence detection was used at an excitation wavelength of 335 nm and an emission wavelength of 440 nm. The separation and quantification of 19 amino acids and 6 amines was carried out in a single run as their OPA/MCE derivatives elute within 80 min, ensuring a reproducible quantification. The method showed to be adequate for the purpose, with an average RSD of 2% for the different amino acids; detection limits varying between 0.71 mg/l (Asn) and 8.26 mg/l (Lys) and recovery rates between 63.0% (Cad) and 98.0% (Asp). The amino acids present at the highest concentration in honey and wine samples were phenylalanine and arginine, respectively. Only residual levels of biogenic amines were detected in the analysed samples.

The characteristics of 59 unifloral honeys of Condalia microphylla Cav. ("piquillín"), Centaurea solstitialis L. ("yellow starthistle") and Prosopis spp., from La Pampa, Argentina, were studied. Pollen features (abundance and frequency of pollen types) and some physicochemical parameters (colour, electrical conductivity, free acidity, glucose content, glucose:water ratio, moisture and pH) were determined. Two different but related sets of calculations were done: the first involved single-factor variance analysis, while the second set involved two multivariate methods, principal component analysis and cluster analysis. Variance and multivariate analysis allowed differentiation of the three honey types according to their physicochemical properties. The variables that best explained this differentiation were pH, electrical conductivity, colour, glucose content and the glucose:water ratio. Pollen analysis showed that the pollen frequency traditionally used (> 45%) for a botanical origin assignment in honey was not valid for the unifloral honeys studied. ("piquillín"), Centaurea solstitialis L. ("abrepuño amarillo") y Prosopis spp., provenientes de La Pampa, Argentina. Se determinaron caracteres políni-cos (abundancia y frecuencia de tipos polínicos) y algunos parámetros fisicoquímicos (color, conductividad eléctrica, acidez libre, contenido de glucosa, relación glucosa-agua, humedad y pH). Se utilizaron dos tipos de análisis estadísticos: análisis de varianza de un factor y análisis multivariados, de componentes principales y de conglomerados. Los análisis de varianza y multivariados permitieron distinguir las tres clases de mieles de acuerdo a las propiedades fisicoquímicas. Las variables que mejor explicaron esta diferenciación fueron pH, conductividad eléctrica, color, glucosa y relación glucosaagua. El análisis polínico demostró que la frecuencia de polen tradicionalmente utilizada para definir una miel monofloral (> 45%) no es válido para las mieles estudiadas.
