Comparison of enzyme-linked immunosorbent assay, radioimmunoassay, complement fixation, anticomplement immunofluorescence and passive haemagglutination techniques for detecting cytomegalovirus IgG antibody.

Abstract: SUMMARY The radioimmunoassay (RIA) and enzyme-linked immunosorbent assay (ELISA) techniques were found to be comparable in sensitivity and specificity for detecting cytomegalovirus IgG antibody, and 10 to 100 times more sensitive than complement-fixation (CF), anticomplement immunofluorescence (ACIF) and passive haemagglutination (PHA). In screening tests for antibody, the frequency of false-positive and -negative results was 0 6% for RIA and ELISA, 1-5% for CF, 1 6% for ACIF and 3 6% for PHA. PHA was the leas… Show more

“…Because of its superior performance, the vast majority of the studies used an enzyme-linked immunosorbent assay (ELISA) to detect CMV-specific IgG. When simply determining whether a person is antibody positive or negative, ELISA is only marginally more sensitive than CF or ACIF [19,20]. However, when measuring titres, the ELISA format is 10-100 times more sensitive than CF or ACIF [20].…”

“…84 Though false negative reactions are rare, and the sensitivity of ELISA for CMV is high, they do occur. [85][86][87] False negative serological reactions may be due to technical reasons, antigenic heterogeneity in CMV, 88 or because some individuals are poor antibody responders. It has been known that more than one-half of CMV-seronegative individuals have CMV DNA.…”
