Comparison of Enzymatic and Microbiological Gentamicin Assays

Abstract: The correlation coefficient between the rapid enzymatic and the ovemight microbiological assays for 211 urine and serum specimens was 0.96. The 95% confidence limits yielded a correlation coefficient between 0.92 and 0.98. Both methods tended to underestimate the amount of a gentamicin added to urine. When only serum samples were considered, the predicted value obtained from the linear regression analysis of either method was within 0.57 ,ug/ml 99% of the time. This high degree of positive correlation will per… Show more

“…Microbiological agar diffusion method was used to estimate the plasma concentration of gentamicin. 22,23 Media (nutrient broth containing 1.5% w/v agar) was added to the sterile culture dishes (90 mm) and allowed to set for 15-20 minutes. S. aureus suspension (equivalent to 0.5 McFarland standard) was spread uniformly onto the surface of the medium with a spreader.…”

“…Results of antibiotic blood levels can be obtained in about 15 min versus 1 to 3 h for radioimmunoassays and radioenzymatic assays, and upwards to 24 h for microbiological assays (5,(7)(8)(9). The time required by the assay is very important if dosage schedules are to be adjusted to achieve optimal therapeutic levels yet avoid toxicity.…”

“…acetyl-CoA) to react with gentamicin yielding a radiolabelled product (gentamicin acetate) which will adhere, by virtue of electrostatic charge to phospho-cellulose paper; the radioactivity can be counted directly (Smith et ai., 1974). The tests, using acetylating and adenylating enzymes, among others, are highly specific and sensitive (Giamarellou et ai., 1975;Smith et al, 1974;Holmes and Sanford, 1974;Daigneault et al, 1974), and by judicious selection of the enzymes, can even be made to discriminate among the various aminogly---cosides (Santanam and Kayser, 1976). Among the problems which have been noted with this kind of assayarehighbackground radioactivity (Holmes and Sanford, 1974), instability and nonuniformityo(fue ingredients (Tilton et al, 1972).…”
