Comparison of different normalization strategies for the analysis of glomerular microRNAs in IgA nephropathy

Abstract: Small nucleolar RNAs (snoRNAs) have been used for normalization in glomerular microRNA (miRNA) quantification without confirmation of validity. Our aim was to identify glomerular reference miRNAs in IgA nephropathy. We compared miRNAs in human paraffin-embedded renal biopsies from patients with cellular-crescentic IgA-GN (n = 5; crescentic IgA-GN) and non-crescentic IgA-GN (n = 5; IgA-GN) to mild interstitial nephritis without glomerular abnormalities (controls, n = 5). Laser-microdissected glomeruli were used… Show more

“…On the other hand, this is the first study that identifies the changes promoted in the serum miRNA profile of SLE patients after treatment with RTX. The selected miRNAs were validated as up‐regulated after RTX treatment included main regulators of inflammation, atherosclerosis, CV disease and nephropathy (ie miR‐28‐5p, miR‐106‐3p and miR‐148b‐3p), as well as inhibitors of IL‐6 production (ie miR‐151a‐3p) . Similar in the case of RA, the changes promoted by RTX therapy in that miRNAs were found parallel to the changes promoted in both disease activity and the inflammatory profile of these patients.…”

“…All differences in sncRNA abundance between the groups that were significant when using spike-in normalization were also significant when using global mean normalization ( S1 Table ). We based our further analysis on the data from the global mean normalization to be as inclusive as possible and because this methodology is considered to be the gold standard [ 23 , 24 ]. When global mean normalization was used, the levels of three sncRNAs were significantly different in pSS patients and nine sncRNAs were significantly different in patients with iSS as compared to HC.…”

“… 47 The mean expression value is often applied for nor-malization 47 but employing only one method could result in missing significant differences. 48 In our study, we normalized our data using the geometric mean of 2 control miRNAs within each group: one snoRNA (SNORD61) and one miRNA (miRNA-211-5p) homogeneously expressed in the discovery set. This approach achieved high concordance across plates and for replicates.…”
