Comparison of Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) and Ciliary Beat Frequency Activation by the CFTR Modulators Genistein, VRT-532, and UCCF-152 in Primary Sinonasal Epithelial Cultures

Abstract: IMPORTANCE Pharmacologic activation of mucociliary clearance (MCC) represents an emerging therapeutic strategy for patients with chronic rhinosinusitis, even in the absence of congenital mutations of the CFTR gene. Drug discovery efforts have identified small molecules that activate the cystic fibrosis transmembrane conductance regulator (CFTR), including potentiators under development for treatment of cystic fibrosis. OBJECTIVE To evaluate the properties of CFTR modulators and their effects on ciliary beat … Show more

“…Stimulation of CFTR-mediated I SC with CFTR potentiators was typically better with primary HSNE as a percentage of overall forskolin-stimulated I SC . 25 Maximal Cl − secretion was augmented with both agents (resveratrol+ivacaftor, 32.2+/−1.17 vs. resveratrol, 10.73+/−0.84 vs. ivacaftor, 21.29+/−0.85 vs. DMSO, 3.88+/−0.85; p<0.001) to levels consistent with what is detected in non-CF HSNE. 2628 …”

“…A high‐speed monochromatic digital video camera (Model A602f‐2; Basler AG, Ahrensburg, Germany) was used to record data at a sampling rate of 100 frames/s. Images were analyzed with Sisson‐Ammons Video Analysis (SAVA, version 2.0.8 W) system . Using the inverted microscope, large regions of beating cilia were identified within HSNE ALI cultures for each experiment.…”
