Comparison of a compatible and an incompatible pepper-tobamovirus interaction by biochemical and non-invasive techniques: Chlorophyll a fluorescence, isothermal calorimetry and FT-Raman spectroscopy

Abstract: imaging technique. PMMoV inoculation also led to a significant rise of ion leakage and heat emission, to the up-regulation of the pepper CCD gene as well as to decreased PSII efficiency, but these responses were much weaker than in the case of ObPV inoculation. Chlorophyll b and total carotenoid contents as measured by spectrophotometric methods were not significantly influenced by any virus inoculations when these pigment contents were calculated on leaf surface basis. On the other hand, near-infrared FTRaman… Show more

“…Following ObPV inoculation the first visible symptoms (hypersensitive necrotic lesions) appeared 72 hpi, in accordance with earlier studies (Rys et al 2014). Pretreatment with 100 nM 24-epi-BR significantly alleviated the visible symptoms of ObPV inoculation (Fig.…”

“…Analysis of chlorophyll (Chl) fluorescence induction curves (slow and fast Chl a fluorescence kinetic) is a very popular method to estimate the effectiveness of photosystems (Papageorgiou et al 2007). This method has been already used during studies devoted to virus-plant interactions (Baron et al 1995, Gonçalves et al 2005, Rys et al 2014. Methods allowing the measurement of leaf reflectance are less popular but also useful for the study of virus-plant interactions, for example for an early detection of viral infection in tomato (Krezhova et al 2014).…”

“…Rys et al [43] compared a compatible (PMMoV) with an incompatible [Obuda pepper virus (ObPV)] peppertobamovirus interaction by Chl-FI and other non-invasive techniques. After the appearance of necrotic lesions, due to inoculation with ObPV, strong inhibition of photochemical energy conversion was observed in these areas; however, leaf tissues adjacent to these inactive areas showed elevated Φ PSII and F V /F M values.…”
