Comparison between the Hybrid Capture II Test and a PCR-Based Human Papillomavirus Detection Method for Diagnosis and Posttreatment Follow-Up of Cervical Intraepithelial Neoplasia

Abstract: Human papillomavirus (HPV) infection is the major cause of cervical cancer and its precursor, cervical intraepithelial neoplasia (CIN), and HPV testing has therefore been proposed for improved triaging and follow-up of women treated for CIN. We compared two common HPV DNA detection tests (Hybrid Capture II [HCII] and PCR-enzyme immunosorbent assay (EIA) using the primers GP5؉/GP6؉ followed by HPV typing with reverse dot blot hybridization) for sensitivity and specificity for detection of CIN and of CIN recurre… Show more

“…HPV DNA detection and genotyping were carried out using the Linear Array (LA) HPV Genotyping Test (Roche Diagnostics). Briefly, HPV DNA was amplified by PCR using a pool of biotin-labeled primers that hybridize in the L1 region (18,23,24 (25). For the purposes of this study, undetermined-risk and LR HPV types were combined.…”

“…k=0.70 before and k=0.72 after the treatment. This suggests that both tests are almost equal [86]. When in cervical smears the approved HC2 test and the nested PCR utilizing the MY/GP primers have been compared, some authors found a higher sensitivity (96.6% versus 58.6%) in favor of the HC2 test [87].…”

“…Moreover, in addition to HPV persistence during follow-up after conization (22) a significant relationship was also found between high HPV load before treatment and recurrent disease (17,24). (25). There are many studies on the clearance of HPV infection after CIN treatment, however the results are quite variable.…”
