Comparison between Immunization Routes of Live Attenuated Salmonella Typhimurium Strains Expressing BCSP31, Omp3b, and SOD of Brucella abortus in Murine Model

Abstract: Live, attenuated Salmonella Typhimurium vaccine candidate expressing BCSP31, Omp3b, and SOD proteins of Brucella abortus was constructed. Thirty BALB/c mice were divided equally into three groups, Group A, were intraperitoneally (IP) inoculated with 100 μl of approximately 1.2 × 106 colony-forming units (CFUs)/ml of the Salmonella containing vector only in 100 μl as a control. And groups B and C mice were orally and IP immunized with approximately 1.2 × 109 CFU/ml of the mixture of three delivery strains in 10… Show more

“…Superoxide dismutase (SOD) forms part of the antioxidant defense system that protects bacteria from the toxic effects of reactive oxygen intermediates by converting superoxide radicals into hydrogen peroxide and oxygen [ 40 ], suggesting that SOD has a crucial role in Brucella intracellular survival. As anticipated, anti- Brucella SOD-based vaccines confer substantial amounts of protection [ 23 52 93 ].…”

“…Recently, our group developed Salmonella Typhimurium delivery-based Brucella vaccines. Using a mouse model, the suitability of the vectored Brucella vaccine in different routes of immunization was studied [ 52 ]. To improve the protective efficacy, we also investigated the usefulness of these vectored vaccines when provided as a “cocktail”.…”

“…Immunoblotting assays in cows immunized with B. abortus S19 also showed an antigen-fighting reaction against antibodies in proteins with a molecular weight of 36 kDa 24 . Antigenic proteins are characterized by their ability to stimulate antibodies in the host's body 23 . In accordance with this statement indicates that the 36 kDa protein has antigenic properties because it can cause antibodies on the host's body.…”

“…In a previous study [ 21 ], S. Typhimurium strains expressing the recombinant B. abortus BCSP31, Omp3b, and SOD proteins were evaluated a vaccine candidate in a murine model. Intraperitoneal immunization with the mixture of the S. Typhimurium - based B. abortus vaccine strains induced adequate humoral and CMI responses and effectively protected against B. abortus infection in mice.…”

“…Especially, the protective efficacy of the S . Typhimurium-based strains against B. abortus infections were mainly evaluated as B. abortus vaccine candidate [ 21 , 24 , 35 , 36 ]. At the first time, the present study aimed to establish a proof-of-concept of the protective efficacy of S. Typhimurium delivery strains expressing recombinant B. abortus BCSP31, Omp3b, and SOD proteins as a B. abortus vaccine candidate in Beagles.…”
