COMPARISON BETWEEN FOUR USUAL METHODS OF IDENTIFICATION OF Candida SPECIES

Abstract: SUMMARYInfection by Candidaspp. is associated with high mortality rates, especially when treatment is not appropriate and/or not immediate. Therefore, it is necessary to correctly identify the genus and species of Candida. The aim of this study was to compare the identification of 89 samples of Candida spp. by the manual methods germ tube test, auxanogram and chromogenic medium in relation to the ID 32C automated method. The concordances between the methods in ascending order, measured by the Kappa index were:… Show more

“…However, as CHROMagar is designed only for the differentiation of three species )i.e., C. albicans [green], C. tropicali s [metallic blue with a pink halo], and C. krusei [pink with velvety appearance]), it fails to clearly identify other species [ 26 ]. Such misclassification of species was also reported in the studies carried out by Estrada et al and Souza et al [ 27 , 28 ].…”

“…C. albicans was cultured for 4 days at 37 • C under aerobic conditions on Malt extract agar made with 4% malt extract (Sladovna Bruntal, Bruntal, Czech Republic) and 1.5% agar (Dr. Kulich Pharma, Hradec Kralove, Czech Republic). Candida tropicalis was obtained from The National Institute of Public Health (NIPH, Prague, Czech Republic); it was isolated from bronchoalveolar lavage and determined by auxanogram [15].…”

“…The germ tube is a small and continuous non-septate elongation of the mother cell that initiates the C. albicansfilamentation. The germ tube's parallel walls do not have a constriction at their point of origin of blastoconidium. It is considered a transitional morphological switch between the yeast and germinating hyphae (32). Candidaalbicans and C. dubliniensisuse the switch as pathogenic properties to increase protein synthesisand ribonucleic acid (33).…”
