The aim of the work was to determine the concentration of serum cystatin C and usability of its measurement in diagnostics of renal failure in cats. The reference point was the value of glomerular filtration rate (GFR), calculated on the basis of plasma clearance of iohexol (Pcio). Seventy cats were divided into two groups: group I -24 healthy cats, group II -46 cats affected with renal dysfunction. The measurement of serum cystatin C was done according to immunonefelometric method. In order to estimate Pcio, the following blood samples were collected: pre-sample -0, and samples collected 120, 180, 240, and 300 min after intravenous administration of iohexol. Its concentration was determined using high performance liquid chromatography. Pcio was obtained using a one-compartment model, according to the method developed by Krutzen Back and Nilsson-Ehl. Serum concentration of cystatin C in cats with renal dysfunction was significantly statistically higher (1.3 ± 0.6 mg/L) as compared to the group of healthy animals (0.7 ± 0.2 mg/L). Statistical analysis of the data confirmed strong correlation between GFR and serum concentration of cystatin C (r = -0.51). Data analysis on the basis of the receiver operating characteristic curve proved that cystatin C seems to be more reliable parameter than creatinine in the assessment of kidneys function in cats and the measurement of cystatin C in blood serum can be of a considerable meaning as far as early diagnosis of renal failure in cats is concerned.

Background: Glomerular filtration rate (GFR) is decreased in humans with hypothyroidism, but information about kidney function in dogs with hypothyroidism is lacking.Hypothesis: Hypothyroidism influences GFR in dogs. The objective of this study was to assess GFR in hypothyroid dogs before implementation of thyroxine supplementation and after re-establishing euthyroidism.Animals: Fourteen hypothyroid dogs without abnormalities on renal ultrasound examination or urinalysis.Methods: Blood pressure and GFR (measured by exogenous creatinine clearance) were measured before treatment (T0, n 5 14) and at 1 month (T1, n 5 14) and at 6 months (T6, n 5 11) after beginning levothyroxine supplementation therapy (20 mg/kg/ d, PO). The response to therapy was monitored at T1 by measuring serum total thyroxine and thyroid stimulating hormone concentrations. If needed, levothyroxine dosage was adjusted and reassessed after 1 month. Statistical analysis was performed using a general linear model. Results are expressed as mean AE standard deviation.Results: At T0, the average age of dogs in the study group was 6.3 AE 1.4 years. Their average body weight decreased from 35 AE 18 kg at T0 to 27 AE 14 kg at T6 (P o .05). All dogs remained normotensive throughout the study. GFR increased significantly with levothyroxine supplementation; the corresponding results were 1.6 AE 0.4 mL/min/kg at T0, 2.1 AE 0.4 at T1, and 2.0 AE 0.4 at T6 (P o .01).Conclusion: GFR was o2 mL/min/kg in untreated hypothyroid dogs. Re-establishment of a euthyroid state increased GFR significantly.

Development and validation of a timed urinary collection system for use in the cat

The aim of the study was to develop and validate a feline urinary collection system for accurate 24 h urine output measurement and glomerular filtration rate (GFR) evaluation. We hypothesized that precise identification of urination time improves the accuracy of the collection system. In a group of nine cats, urinary volume and micturition times were repeatedly recorded for up to 48 h using purpose-built collection trays containing a temperature data logger. Collection time was determined both with and without using the data loggers on 22 occasions and agreement between estimated 24 h urine outputs obtained with the two calculation methods was evaluated. GFR was repeatedly measured by endogenous urinary creatinine clearance on three occasions. Twenty-four-hour urine output was measured in 98.5% of the attempted collections (300 cat-sampling days). Sensitivity and specificity of the detection system were 97.8% and 100%, respectively. Mean 24 h urine output was 12.4 + 4.94 mL/kg/day and mean intra-cat between-days coefficient of variation (CV) was 16.6 + 5.6% when data loggers were used. The absolute relative volume error between the two calculation methods ranged from 0% to 131%. Median absolute relative [interquartile range] error was 9.1% [3.25-19.8]. Bias was 21.3% and lower and upper limits of agreement were 239.7% and 35.2%, respectively. Mean estimated GFR was lower than previously reported with comparable urinary clearance methods (1.92 + 0.37 mL/min/kg) and mean within-cat CV was 12 + 6.9%. The system was simple in design, readily affordable, allowed normal micturition behaviour and reduced intra-animal variability in 24 h feline urine collection.
