Comparision of the Inotropic Effects of Levosimendan, Rolipram, and Dobutamine on Human Atrial Trabeculae

Abstract: The aim of this study was to compare the positive inotropic effects of 3 different agents with 3 different mechanisms of actions-levosimendan, rolipram, and dobutamine-on human atrial trabecular muscles. Samples of right atrial appendage (1 cm, 500-1000 mg) were removed and immersed in preoxygenated and modified Tyrode solution. In oxygenated Tyrode solution, preparations were used to investigate the concentration-effect relationship of levosimendan, dobutamine, and rolipram on percentage developed tension (DT… Show more

“…4 were performed to confirm that the effect of 5-HD on RPP was due specifically to inhibition of contractility. Superfusion of human atrial trabeculae in vitro with dobutamine increased contractility to 161 Ϯ 32% of baseline, as is typical of this preparation (29). 5-HD caused a 61% reduction in the dobutamine response, and 1 M glyburide caused a 30 Ϯ 3% (n ϭ 3 experiments) reduction in the dobutamine response (data not shown).…”

“…Samples of right atria appendages were obtained during cannulation for cardiac surgery. The methods to obtain human atrial trabeculae working isometrically in vitro have been previously described in detail (3,9,29). Briefly, a sample of the right atrial appendage (1 cm 2 , 500 -1,000 mg) was removed and immersed in preoxygenated and modified Tyrode solution containing (in mM) 120 NaCl, 4 KCl, 2.7 CaCl 2, 1.1 MgCl2, 25.7 NaHCO3, 1.8 NaH2PO4, and 11 glucose at 22°C.…”

“…18 It is well known that levosimendan increases Ca +2 sensitivity of the heart by myofibrillar Ca +2 sensitization via Ca +2 -dependent stabilization of the Ca +2 -bound conformation of cardiac troponin C 1,20,21 without affecting the Ca +2 affinity of troponin C. Phosphodiesterase inhibition may also play a role in the cardiac effect of levosimendan. 22,23 Accordingly, it has been noted that the activity of phosphodiesterases may prevent intracellular cAMP accumulation following levosimendan application in both human and guinea-pig myocardium.…”

“…The methods to obtain human atrial trabeculae working isometrically in vitro have been previously described in detail. [17][18][19] Briefly, a sample of the right atrial appendage (1 cm 2 , 500-1000 mg) was removed and immersed in preoxygenated and modified Tyrode's solution (in mM: NaCl, 120; KCl, 4; CaCl 2 , 2.7; MgCl 2 , 1.1; NaHCO 3 , 25.7; Na H 2 PO 4 , 1.8; and glucose, 11) at 22 ºC. The time between excision and beginning of laboratory processing was 1-5 min.…”
