Comparing bacterial membrane interactions and antimicrobial activity of porcine lactoferricin-derived peptides

Abstract: Antibiotic treatment for microbial infections is under scrutiny due to increasing resistance to conventional antibiotics, warranting discovery of new classes of antibiotic agents. Antimicrobial peptides are part of the innate defense system found in nearly all organisms and possess bactericidal mechanisms that make it more difficult for bacteria to develop resistance. Porcine lactoferricin (LFP-20) is an antimicrobial peptide located in the N terminus of lactoferrin (LF). To develop novel cell-selective antimi… Show more

“…The outer membrane (OM) permeability activity against Gram-negative bacteria was determined by the uptake of N -phenyl-1-naphthylamine (NPN), a fluorescent dye that is sensitive to the outer membrane, as described previously [14,48]. Briefly, E. coli UB1005 grown to the logarithmic phase (OD600 = 0.4) at 37 °C in MHB were harvested by centrifugation at 1000× g for 10 min at 4 °C.…”

“…The effects of Lfcin4 and Lfcin5 on the outer membrane of S. aureus ATCC25923 and S. enteritidis CVCC3377 were determined by measuring the uptake of NPN4445. Mid-log phase cells were washed with sterile normal saline three times and suspended in 5 mM HEPES buffer (pH 7.2, containing 5 mM glucose) to an OD at 600 nm of 0.5.…”

“…Lfcin B is a 25-residue peptide fragment discharged from the N-terminal domain of bovine lactoferrin (residues 17~41) upon pepsin digestion, bovine lactoferrin (LF) is an 80 kDa iron-binding glycoprotein that is found in the secretory granules of neutrophils and in biological fluids, including saliva and milk. Lfcin B is a cationic antimicrobial peptide with cytotoxic activity against microorganisms [1]. It consists of 25 amino acid residues (17 to 41 proximal to the NH2 terminus of bovine lactoferrin) and is produced by acid-pepsin hydrolysis of bovine lactoferrin.…”
