Comparative Transcriptomic Analysis Uncovers Genes Responsible for the DHA Enhancement in the Mutant Aurantiochytrium sp.

Abstract: Docosahexaenoic acid (DHA), a n-3 long-chain polyunsaturated fatty acid, is critical for physiological activities of the human body. Marine eukaryote Aurantiochytrium sp. is considered a promising source for DHA production. Mutational studies have shown that ultraviolet (UV) irradiation (50 W, 30 s) could be utilized as a breeding strategy for obtaining high-yield DHA-producing Aurantiochytrium sp. After UV irradiation (50 W, 30 s), the mutant strain X2 which shows enhanced lipid (1.79-fold, 1417.37 mg/L) and … Show more

“…DHA is a n-3 long-chain polyunsaturated fatty acid and is critical for cellular processes involved in maintaining health. In the study of Liu et al (2020) [13], UV mutagenesis was utilized to obtain competitive Aurantiochytrium sp. strain with enhanced DHA production (1.90-fold higher than wild strain).…”

“…The sequence of Aurantiochytrium sp. SZU445 18s rDNA (GenBank: MT232522.1) (Liu et al, 2020;Zhu et al, 2020) was amplified with the primer pair 18S-F/18S-R. The sequence of DECR was amplified from Aurantiochytrium sp.…”

“…To date, transcriptomic studies have been conducted on A. limacinum and other thraustochytrids (Dellero et al, 2020; Heggeset et al, 2019; Liang et al, 2018; Liu et al, 2020; Ma et al, 2015; Ye et al, 2019). However, most of these studies have primarily focused on the transcriptional profiles of genes related to lipid or PUFA biosynthesis pathways (Heggeset et al, 2019; Liang et al, 2018; Liu et al, 2020; Ma et al, 2015). Therefore, we are still far from a complete understanding of how A. limacinum regulates the production of astaxanthin and other carotenoids (Ye et al, 2019).…”
