Comparative transcriptome analysis of flower heterosis in two soybean F1 hybrids by RNA-seq

Abstract: Heterosis has been widely exploited as an approach to enhance crop traits during breeding. However, its underlying molecular genetic mechanisms remain unclear. Recent advances in RNA sequencing technology (RNA-seq) have provided an opportunity to conduct transcriptome profiling for heterosis studies. We used RNA-seq to analyze the flower transcriptomes of two F1 hybrid soybeans (HYBSOY-1 and HYBSOY-5) and their parents. More than 385 million high-quality reads were generated and aligned against the soybean ref… Show more

“…The total RNA was extracted using TRIzol (Thermo Scientific TM). cDNA was synthesized using PrimeScript RT Reagent Kit (TaKaRa, Dalian, China) . A 5 μL PCR mixture consisted of 2.5 μL of SYBR mixture (TaKaRa), 0.2 μL of cDNA, 0.2 μL of each PCR primer, and 2.1 μL of ddH 2 O.…”

“…Plant secondary metabolites are crucial for the plant in adapting to an adverse environment for defense [31]. A large number of DEGs were assigned to the pathway related to various secondary metabolite syntheses, indicating their role in making the hybrids better-suited to the environment [11,32]. Similarly, early light-induced protein (ELIP) and chloroplast precursor genes were also highly up-regulated in both the hybrids at the GF stage.…”

“…Acquiring such information through transcriptome-wide methods has recently gained traction in understanding gene expression basis of heterosis. Using microarray or RNA-Seq, heterosis has been analyzed in species such as rice [9], maize [10], soybeans [11], and silkworm [12]. However, to the best of our knowledge, there is no published report on gene expression analysis of heterosis in chickens or in any other livestock species.…”
