Comparative studies of the toxicity of standard reference materials in various cytotoxicity tests and in vivo implantation tests

Abstract: Polyurethane films that contained various amounts of zinc diethyldithiocarbamate (ZDEC) and zinc dibutyldithiocarbamate (ZDBC) were prepared as standard reference materials (SRM). Using three cell lines of V79, L929, and Balb/3T3 cells, the cytotoxicity of the dithiocarbamates and the SRM films were compared by agar diffusion assay, filter diffusion assay, neutral red assay, cell growth assay, and colony assay. Among these in vitro cytotoxicity tests, colony assay was found to be the most sensitive method fo… Show more

“…The use of ZCF as cytotoxic control has been established previously for multiple cell types with varying cytotoxic responses 20,21,31,32 . L929 cells had the greatest cytotoxic response to ZCF when compared to the MC3T3, hMSC and cMSC cells.…”

“…Established cell lines are preferred and can be obtained from recognized repositories. Where a specific sensitivity is required, primary cell cultures and cell lines obtained directly from living tissues should only be used if reproducibility and accuracy of the response can be demonstrated 14-16. However, several problems, such as the preparation of test materials (in the case of the extract method), choice of cell type, the test procedure and method used to quantify results, etc., have been encountered in vitro cytotoxicity tests 17-19.…”

“…The reported results were in accordance with ours, even if the cell lines are different, they are both mice fibroblasts widely used in cytotoxicity tests. Tsuchiya et al . performed a comparative study of the toxicity of standard reference biomaterials using both L929 and Balb 3T3 cells and concluded that at the neutral red uptake assay, Balb 3 T3 presents higher sensitivity than L929 cells.…”
