Comparative proteomics of the two T. brucei PABPs suggests that PABP2 controls bulk mRNA

Abstract: Poly(A)-binding proteins (PABPs) regulate mRNA fate by controlling stability and translation through interactions with both the poly(A) tail and eIF4F complex. Many organisms have several paralogs of PABPs and eIF4F complex components and it is likely that different eIF4F/PABP complex combinations regulate distinct sets of mRNAs. Trypanosomes have five eIF4G paralogs, six of eIF4E and two PABPs, PABP1 and PABP2. Under starvation, polysomes dissociate and the majority of mRNAs, most translation initiation facto… Show more

“…Although PBP1 interacts with both PABP1 and PABP2 in the yeast 2-hybrid assay (25), only PABP2 copurified with XAC1. This result is consistent with a published study: PBP1 was enriched in pull-downs of PABP2, but not PABP1 (70). Procyclic-form PABP2 pull-downs, especially under low-salt conditions, also contained the MKT1 complex and several MKT1-interacting proteins, including ZC3H20, ZC3H28, ZC3H39, PUF6 and PUF9.…”

“…In T. brucei , PABP2 may accumulate in nuclear periphery granules, most likely regulating the nuclear export of transcripts [40]. TbDRBD2 is also present in the TbPABP2 complex, together with TbZC3h40, TbALBA1, and TbALBA3 [41]; these results are consistent with ours (Tables 1 and 2).…”

“…RBP42, which is a polysomeassociated protein that preferentially binds to coding regions (24), also preferred TUB. Only one of the two poly(A) binding protein orthologues, PABP2, was reproducibly associated with either VSG or TUB mRNAs, supporting previously suggested (25) specialized roles for PABP2 and its orthologue PABP1.…”
