Comparative proteomics of inhaled silver nanoparticles in healthy and allergen provoked mice

Abstract: Background: Silver nanoparticles (AgNPs) have been associated with the exacerbation of asthma; however, the immunological basis for the adjuvant effects of AgNPs is not well understood. Objective: The aim of the study reported here was to investigate the allergic effects of AgNP inhalation using proteomic approaches. Methods: Allergen provoked mice were exposed to 33 nm AgNPs at 3.3 mg/m 3 . Following this, bronchoalveolar lavage fluid (BALF) and plasma were collected to determine protein profiles. Results: In… Show more

“…Some more recent in vivo health studies are available, but of these, only a dozen use low dose rate inhalation as the method of Ag NP administration (Hyun et al , 2008, Sung et al , 2008, Sung et al , 2009, Stebounova et al , 2011, Sung et al , 2011, Kwon et al , 2012, Cho et al , 2013, Chuang et al , 2013, Roberts et al , 2013, Song et al , 2013, Su et al , 2013, Braakhuis et al , 2014). Route and method of delivery can have significant effects on physicochemical particle characteristics and physiological responses (U.S.…”

“…Inhalation studies with uncoated Ag NPs suggest that they produce inflammation, allergic reactions, decrements in lung function, and/or DNA damage (Sung et al , 2008, Sung et al , 2009, Kwon et al , 2012, Cho et al , 2013, Chuang et al , 2013, Su et al , 2013, Song et al , 2013, Braakhuis et al , 2014) following acute, sub-chronic, and/or chronic exposures. In contrast, numerous other inhalation studies suggest that uncoated Ag NPs produce no adverse (toxic) responses (Hyun et al , 2008, Stebounova et al , 2011, Sung et al , 2011, Roberts et al , 2013).…”

“…For instance, protein abundances of the phosphorylation-based p70S6K signaling pathway and protein ubiquitination pathway were reported to be altered with selenium quantum dots treatment [62]. In addition, proteins involved in ubiquitination were found altered in human lung cells treated with copper-oxide ENMs [61], Balb/3T3 mouse fibroblast cells treated with gold ENMs [35], and mice that inhaled silver ENMs [63]; however, the identity of modified proteins has not been reported.…”
