Comparative Proteome Analysis of Multi-Layer Cocoon of the Silkworm, Bombyx mori

Abstract: Bombyx mori cocoon has a multi-layer structure that provides optimal protection for silkworm pupa. Research on the mechanical properties of the multi-layer structure revealed structure-property relationships of the cocoon. Here, we investigated the protein components of the B. mori cocoon in terms of its multi-layer structure. Liquid chromatography-tandem mass spectrometry identified 286 proteins from the multiple cocoon layers. In addition to fibroins and sericins, we identified abundant protease inhibitors, … Show more

“…3, designated as spot 5Ј), but we suspect that this spot was some other protein(s) as Fhxh4/ Fhxh5 transcripts were not detected in normal PSGs (see the description later). Fhxh4/Fhxh5 proteins are abundant in the cocoon of normal individuals (37) and are predicted to be major silk proteins similar to sericins.…”

“…The composition of silk fibroin is almost consistent from the innermost to the outermost layers of the silkworm cocoon [101]. Silk sericin is considered as a bonding protein among hydrophilic silk fibers.…”

“…The sericin in the innermost layer is more or less 15%, and in the outermost layer, it is 25% [102]. Seroin is considered to be an anti-septic protein, and its content decreases from the inner to the outer layers [101]. The amount of various protease inhibitors is also different according to the layer [103].…”
