Comparative Proteome Analysis of Breast Cancer and Adjacent Normal Breast Tissues in Human

Abstract: Two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) and matrix-assisted laser desorption/ionization tandem time-of-flight mass spectrometry (MALDI-TOF/TOF-MS), incorporated with online database searching, were performed to investigate differential proteins of breast cancer and adjacent normal breast tissues. Considering that serum albumin is abundantly presented in normal control samples, 15 differential spots detected in 11 out of 12 (91.7%) breast cancer samples were identified by online SIENA-2DPAG… Show more

“…these proteins involved stress response, apoptosis, protein synthesis/degradation, homeostasis, cell migration and immune defense. some of the proteins have been reported in previous study comparing breast tumor and normal tissue (10)(11)(12). the results from 2D-sDs-pAge showed a very similar protein expression profiles in primary tumor and matched Aln and between tumors with and without Aln metastases.…”

“…Because of its anti-apoptotic properties, it is a key pro-survival component of the unfolded protein response (17). HspA5 has been reported to be up-regulated in breast cancer compared to normal tissue (10,11,18). In the present study, HspA5 was up-regulated in metastatic Aln, supporting the theory that HspA5 plays a role in the protection of metastatic cells in new adverse environments.…”

“…TCTP was overexpressed in many human cancer tissues including liver (42), colorectal (43), prostate (44), breast (45), and lung cancer (46). Similarly, TCTP was detected in pancreatic cancer cell lines, and malignant duct epithelial cells and some normal acinar cells but not normal pancreatic duct epithelial cells adjacent to tumor foci.…”

“…It is reported as multifunctional to participate in numerous cellular processes including cell growth, survival, development, protein synthesis, immune response, tumor reversion, transcription regulation, and induction of pluripotent stem cells or apoptosis (Bommer and Thiele 2004). Comparing to normal tissue, the level of TCTP is increased in many human cancer tissues, such as breast, colorectal, prostate and lung cancer (Arcuri et al 2004;Deng et al 2006;Kim et al 2008;Slaby et al 2009). Therefore, in recent years, more and more attention is paid to the involvement of TCTP on tumor reversion because of its critical role in tumorigenesis (Miao et al 2013;Nagano-Ito and Ichikawa 2012).…”
