Comparative physicochemical stability and efficacy study of lipoid S75-biopeptides nanoliposome composite produced by conventional and direct heating methods

Abstract: Liposomes are used as effective nanodelivery devices to improve the physicochemical stability and biological efficacy of the encapsulated peptides and proteins. In this study, nanoliposome composite of lipoid S75entrapped angiotensin I-converting enzyme (ACE)-inhibitory biopeptides was prepared by conventional (BLS75-CM) and direct heating (BLS75-DHM) methods. The nanoliposomes (BLS75-CM and BLS75-DHM) were stored at 4°C for 8 weeks and evaluated for physicochemical stability in terms of particle size, polydis… Show more

“…A 200 µL each of bromelain-and papain-generated DCB protein hydrolysates were separately mixed with 200 µL of 20% (w/v) TCA solution and allowed to stand for 30 min. The precipitate formed was centrifuged at 7800 x g for 12 min and the TCA-soluble peptide content of the resulting supernatant was determined using BCA protein assay kit method as previously reported (Auwal et al, 2018a;Auwal et al, 2018b) with bovine serum albumin (BSA) as standard. The TCA-soluble peptide yield was calculated as follows: TCA -soluble peptide (%) = X/Y x 100% where X: peptide content of the supernatant and Y: total protein content of the sample before enzymatic hydrolysis.…”

“…Besides, results of the in vivo efficacy of these chitosan nanoparticles in SHRs indicated a dose-dependent blood pressure lowering effect that was significantly higher than that caused by the non-encapsulated peptides [111]. In addition, the encapsulation of stone fish-derived bioactive peptides in nanoliposomes improved peptide stability against in vitro gastrointestinal digestion, although no data about the antihypertensive effect were provided [112]. Similarly, improved stability and bioavailability of nanoliposomes loaded with an ACE inhibitory peanut peptide fraction in a simulated gastrointestinal tract were reported [108].…”
