“…Since the positions of rDNA sequences provide useful markers for chromosome identification, physical mapping of rDNA has been reported in many plant species including bryophyte (Fukui et al 1994, Lubaret et al 1996, Fukui et al 1998, Roose et al 1998, Cerban et al 1999, Moscone et al 1999, Taketa et al 1999, Sone et al 1999, Siroky et al 2001, Tagashira and Kondo 2001, Shibata et al 2004. Although the locations of 5S rDNA loci differ from the locations of 45S rDNA loci in most plant species, the 5S rDNA and 45S rDNA loci in several other plant species are located in close proximity (Roose et al 1998, Taketa et al 1999.…”

“…For instance, Trifolium genomic reorganization may occur during speciation and explain species-specific distribution patterns of 5S and 18S (Ansari et al 1999). There is other evidence of the large amount of variation in the numbers and locations of ribosomal genes between closely related species in certain genera (Taketa et al 1999;Adams et al 2000), between different individuals in some species (Frello and HeslopHarrison 2000;Luo et al 2006), and even within individual plants in Allium (Schubert and Wobus 1985), suggesting that the number and location of rDNA can change readily.…”

“…rDNA sequences are also useful chromosome landmarks and provide important information about genome evolution at the chromosomal level. In some species, such as Arabidopsis thaliana [7], Hordeum vulgare [8] and Rhoeo spathacea [9,10], fluorescence in situ hybridisation (FISH) with 25S and 5S rDNA as probes is sufficient to identify each chromosome in the complement. A complementary approach that can be used for karyotype characterization is double fluorescent banding with DAPI and CMA fluorochromes, which discriminates between ATand GC-rich DNA regions.…”
