Comparative phyloproteomics identifies conserved plasmodesmal proteins

Abstract: Plasmodesmata are cytosolic bridges, lined by the plasma membrane and traversed by endoplasmic reticulum; plasmodesmata connect cells and tissues, and are critical for many aspects of plant biology. While plasmodesmata are notoriously difficult to extract, tissue fractionation and proteomic analyses can yield valuable knowledge of their composition. Here we have generated two novel proteomes to expand tissue and taxonomic representation of plasmodesmata: one from mature Arabidopsis leaves and one from the moss… Show more

“…This points to a functional diversification of PD and non-PD-localized members of the GHL17 family or that only PD-localized PpGHL17s can degrade callose at PD. In recent parallel work, GHL17_4 (Pp3c11_25840) and GHL17_18 (Ppc16_15860), also members of the GHL17 alpha clade, were localized to PD in P. patens (Johnston et al, 2023), thus adding further evidence for PD localization of alpha clade GHL17s.…”

“…Their meristems have a simpler organization while at the same time still sharing genetic control elements with higher plants [ 141 ]. Furthermore, knowledge of the molecular makeup of bryophyte plasmodesmata is emerging, along with tools for studying plasmodesmal function [ 67 , 142 , 143 ]. Beyond the advantages in terms of a more accessible body plan, the inclusion of bryophytes could shed an evo-devo light on the control mechanisms for mobile developmental regulators.…”

“…Therefore, live-cell, light-based fluorescence microscopy with higher resolving power in combination with markers that distinguish plasmodesmal morphology type could be a promising approach. The expanding knowledge of protein presence at different plasmodesmata via proteomics could provide candidates for such markers [ 67 ].…”

“…S11 ). Irrespective of the working model, PDLPs are only found in seed plants ( 28 – 32 ), while NHL-related proteins have orthologues the earlier land plant Physcomitrium patens ( 30 , 33 ). This allows speculation that PDLPs were co-opted into a preexisting complex of NHLs that activates callose synthases to enable an extra regulatory mechanism for plasmodesmata responses.…”
