Comparative pharmacokinetics and metabolites study of seven major bioactive components of Shaoyao‐Gancao decoction in normal and polycystic ovary syndrome rats by ultra high pressure liquid chromatography with tandem mass spectrometry

Abstract: A simple and sensitive liquid chromatography with tandem mass spectrometry method was developed for simultaneous quantification of paeoniflorin, albiflorin, oxypaeoniflorin, liquiritin, liquiritigenin, glycyrrhetinic acid, and glycyrrhizin in rat plasma after oral administration of Shaoyao-Gancao decoction, which is traditionally used in the treatment of polycystic ovary syndrome. The plasma samples were pretreated with methanol as precipitant. The method exhibited good linearity (correlation coefficient (R 2 … Show more

“…After 1°week of acclimatization, the PCOS rats model was induced as described previously ( Liu et al, 2019 ). In brief, animals were orally administrated daily with 1 mg/kg letrozole for 21 consecutive days.…”

“…Intestinal carboxylesterase is a critical enzyme that generated benzoic acid from albiflorin. Benzoic acid is a key intermediate metabolite that crosses the blood–brain barrier, improves brain function, and exerts antidepressant activity in the body (Liu et al, 2019). M36 displayed [M–H] − ion at m/z 137.0239, 16 Da more than [benzoic acid–H] − ion, suggesting that the metabolite was generated from the hydroxylation of benzoic acid.…”

“…The typical product ions at m/z 121.0284 ([M–H–O] − ) and 93.0346 ([M–H–CO 2 ] − ) were detected. M36 was elucidated as m ‐hydroxybenzoic acid (Liu et al, 2019). Besides, after the elimination of the glucose, several other metabolic reactions were apt to be happened due to the hydroxyl group.…”

“…It is shown that the ability of gastric emptying was reduced in diabetes mellitus pathological condition, resulting in a significant increase in the absorption (Gong et al, 2015). In addition, many kinds of drug metabolizing enzymes and P-glycoprotein, especially the cytochrome P450, changed significantly in diabetic rats compared with normal rats, subsequently influencing the metabolism and elimination processes of the compounds (Darakjian et al, 2021;Liu et al, 2019).…”

“…However, to the best of our knowledge, there were fewer reports about the quantification and pharmacokinetic study of SC lignans in rat plasma, except the determination of manassantin B in rat plasma using HPLC with fluorescence detection (Lee et al, 2016) and our previously developed liquid chromatography with tandem mass spectrometric detection (LC-MS) method for the quantitative analysis of sauchinone in rat plasma (Xu et al, 2012). Moreover, there were fewer reports about their pharmacokinetic profiles in disease model rats whose pharmacokinetic process was usually different from those in normal rats (Bai et al, 2019;Liu et al, 2019).…”
