Comparative Genomics of Acinetobacter baumannii Clinical Strains From Brazil Reveals Polyclonal Dissemination and Selective Exchange of Mobile Genetic Elements Associated With Resistance Genes

Abstract: Sequencing Multiple Brazilian Acinetobacter Genomes genes associated with the synthesis of the capsular antigens were noticeably more variable in the ST113 and ST79 strains. Indeed, several resistance and virulence genes were common to the ST79 and ST113 strains only, despite a greater genetic distance between them, suggesting common means of genetic exchange. Our comparative analysis reveals the spread of multiple STs and the genomic plasticity of A. baumannii from different hospitals in a single metropolitan… Show more

“…The results show that the number of ARGs does not correlate with the number of ISs in the four species (Pearson’s R2 < 0.8, Figure 2). Previously, it was reported that at least eight MGEs were detected together with ARGs in A. baumannii (Leal et al, 2020). The plasmids were grouped into three categories based on the DNA transfer machinery: conjugative, mobilizable and nonmobilizable (Smillie et al, 2010).…”

“…The most prevalent carbapenemase in A. baumannii strains, OXA-23, was detected here in already associated clones as ST1, ST25 and ST79 ( 23 , 30 ). Although this gene was not detected in some ST79 strains from this surveillance, all of them have OXA-65 carbapenemase, as previously reported in a Brazilian surveillance ( 31 ). The single strain from A. baumannii ST848 calls to attention since the occurrence of bla GES−11 , which mediates resistance to β-lactam and reduced susceptibility to carbapenem, has been reported in other continents and clones but neither in America nor in ST848 ( 32 ).…”

“…However, there is a second waaaL gene in the A. baumannii was found in some strains, which thought to be required in O-antigen linkage as it does not contain a domain for protein glycosylation [ 69 , 70 ]. In contrast to previous study, recent comparative genomics of A. baumannii in brazil has suggested no waaL was identified in the strains using blastp and Pfam for gene identification, and Artemis compare gene arrangement [ 71 ].…”
