The purpose of the present work was to study effectiveness for early pregnancy diagnosis in cattle of the new enzyme immunoassay (EIA) sandwich kit commercially available based on the measurement of pregnancy-associated glycoproteins (PAGs). 120 Holstein-Friesian cattle of mixed age and parity were comprised from different dairy herds. The pregnant females (n = 68) were diagnosed by ultrasonography at day 35-40 after artificial insemination and confirmed by transrectal exploration at 2-3 months after AI. The non-pregnant females (n = 52) were housed in the absence of males during the experimental period. Blood samples were collected from coccygeal vessels of females into EDTA tubes. The serum was obtained by centrifugation and the serum was stored at - 20 C until assay. The PAG concentrations in pregnant and non-pregnant females were determined in serum by EIA kit. The reproducibility inter- and intra-assay of the PAG-EIA is satisfactory (2.78 and 13.19 %, respectively). The accuracy ( 94.8 %) and the test of parallelism were largely acceptable. No cross-reaction was observed with the different hormones tested at different dilutions. PAG-EIA system gave 100 % sensitivity and negative predictive values. Whereas, specificity and positive predictive value were 91.93 and 71.15 %, respectively. The accuracy of pregnancy diagnosis by PAG-EIA was 87.5 %. In conclusion, the present study shows clearly that the EIA kit can be used to measure PAG in serum cows for the detection of gestation in Algeria. Therefore, this alternative technique could be recommended to replace the radioactive methods in immunoassays to improve the reproductive performances and an efficient tool for reproductive management of dairy cattle.

Background Placenta-specific protein 1 (PLAC1) is a small secreted protein considered to be a molecule with a significant role in the development of the placenta and the establishment of the mother-foetus interface. This study aimed to confirm the presence of bovine PLAC1 and to examine its profile in the placenta and plasma in the first six months of pregnancy. The expression pattern of PLAC1 was analysed by RT-qPCR and Western Blotting. Quantitative evaluation was carried out using ELISA. Results PLAC1 concentrations in the plasma of pregnant cows were significantly higher (p < 0.05) than those obtained from non-pregnant animals. PLAC1 protein concentrations in the placental tissues of the foetal part were significantly (p < 0.05) higher than in the tissues of the maternal part of the placenta. PLAC1 transcripts were detected in both placental tissue samples and epithelial cell cultures. Conclusions In conclusion, the results of the present preliminary study suggest that PLAC1 is involved in the development of bovine placenta. The presence of this protein in the plasma of pregnant animals as early as the first month may make it a potential candidate as a pregnancy marker in cows. Further studies on exact mechanisms of action of PLAC1 in bovine placenta are necessary.

INTRODUCTION: With the constant change in information technology, people have entered the era of big data, and the reforms brought about by the era of big data have profoundly affected people's way of life and learning. English writing is one of the four basic skills for mastering English, and the skilful mastery of writing is an essential form of accurate expression in English. Different from the traditional teaching modes of "separate assessment of teaching" and "separate assessment of learning", "cooperative assessment between teachers and students" is a new type of assessment mode proposed by professors at the National University in recent years. This cooperative evaluation model advocates the combination of the teaching and evaluation processes and the evaluation and learning processes. In the cooperative evaluation model, evaluation is considered an extension of the teaching process, and the evaluation process itself is also a school process. The arrival of the significant data era also brings new opportunities and challenges for university English teacher-student cooperative evaluation. OBJECTIVES: We design a university English writing teacher-student cooperative evaluation system based on big data by combining big data with the teacher-student cooperative evaluation. METHODS: All college English writing classes in one university were selected as the background for ample data research, and with the help of theoretical knowledge and technology related to big data, a model was constructed to analyze the factors affecting the cooperative evaluation system of college English writing teachers and students. RESULTS: The university English writing teacher-student cooperation evaluation system is analyzed and summarized using big data. CONCLUSION: By using big data to analyze the evaluation of college English writing teacher-student cooperation, big data can better help teachers understand the weaknesses of students' knowledge points and better help students improve their English proficiency.
