Chapman's medium designed for the enumeration of Staphylococcus aureus showed three shortcomings: (i) micrococci and many Bacillus spp. grow well on this agar and may obscure colonies of Staph. aureus; (ii) some strains of Staph. aureus fail t o dissimilate mannitol; among 1164 strains examined, mostly from clinical sources, 4.4 yo appeared unable to dissimilate mannitol; (iii) the agar has inhibitory properties to sublethally impaired cells of Staph. uureus. The latter deficiency not being correctable, the suitability of the egg yolk-tellurite-glycine-pyruvate agar (ETGPA) of Baird-Parker (1962b) was evaluated. Of 522 strains of Staph. aureus of human and food origin 97.5 yo developed characteristically and generally quantitatively on Baird-Parker's agar.Raird-Parker's medium has proved valuable in ecological studies on the occurrence and development of Staph. aureus in foods incriminated in staphyloenterotoxicosis : among 165 atypical black colonies isolated from 450 food samples only 2% were Staph. aureua.

Baird‐Parker (BP), modified BP medium (egg yolk replaced with pig plasma, BPP) and modified Vogel and Johnson agar (phosphatidyl choline, deoxyribonucleic acid, with catalase added, PCVJ) were equally efficient for enumerating both non‐stressed and thermally‐stressed populations of Staphylococcus aureus from pure cultures and naturally contaminated foods. Vogel and Johnson agar was inferior. All colonies exhibiting coagulase activity on BPP were subsequently confirmed as Staph. aureus; this was not the case with presumptive colonies from BP and PCVJ. Based on selectivity, definitive diagnostic characterization of colonies and increased sensitivity (more sample plated), BPP should be considered as the reference method for the routine enumeration of Staph. aureus in foods.

Summary: A direct plating procedure was used in 7 laboratories for detecting and enumerating coagulase positive staphylococci in 424 foods and for the recovery of staphylococci from 233 foods experimentally inoculated in the laboratory. Coagulase positive staphylococci were isolated from 46 (10·9%) of the 424 routine foods. For general use a reliable and selective medium is required which is inexpensive, simple to prepare and stable on storage. Eleven selective media were tested and of these phenolphthalein‐diphosphate agar with polymyxin, milk‐ or cream‐salt agar and mannitol‐salt agar were considered to be the most useful.
