Comparative Estimates of Bacterial Affinities and Adsorption Sites on Hydroxyapatite Surfaces

Abstract: The adsorption of strains of prominent oral bacteria to hydroxyapatite (HA) surfaces was studied by use of an adsorption model based on the Langmuir adsorption isotherm; this permitted comparative estimates of the number of adsorption sites and the strength of the adsorption bonds on untreated and salivatreated HA surfaces for strain of Streptococcus mutans, S. salivarius, S. sanguis, S. mitis, Actinomyces viscosus , and A. naeslundii . The experimental data clos… Show more

“…Previous research has demonstrated the ability of A. viseosusT\'^\ to adhere to saliva-coated hydroxyapatite beads (4,14,18). In the present study, we used hydroxyapatite disks because the bubble activity and streaming effects of the acoustic energy on the fiuid made use of beads impractical.…”

“…In order to examine the effect of IgAl protease on the adherence-inhibiting activity of SIgA antibody, the hydroxyapatite (HA) adherence assay described by Clark et al (2) and modified by Kilian et al (12) and others (10,21) was employed. Aliquots of 40 mg of HA beads (BDH Chemicals, Ltd., Poole, England) were placed in 1.8 ml polypropylene microcentrifuge tubes and washed once with buffered KC1 to remove "fines" and allowed to equilibrate in 1.6 ml of buffered KC1 overnight.…”

“…Hydroxyapatite adsorption assay A modified version of the hydroxyapatite-bacterial adsorption assay, which has been described previously (11), was used to compare the in vitro adsorption affinities, of the Actinomyces strains used in these experiments. Briefly, Actinomyces strains were cultured in Tryptic Soy Broth supplemented with 10 |iCi of [^H] thymidine per ml (Amersham, Arlington Heights, IL).…”

“…Whole human and BALB/c mouse saliva were collected and stored until used as previously described (11,16). Hydroxyapatite preparations treated with human saliva or BALB/c mouse saliva were prepared by incubating 5 mg of washed spheroidal hydroxyapatite beads (BDH Biochemicals Ltd., Poole, UK) in 200 \i\ of whole human or mouse saliva for 1 h as previously described (16).…”
