Comparative Cytopathology and Immunocytochemistry of Japanese, Australian and Brazilian Isolates of Orchid fleck virus

Abstract: Cytopathic effects in orchid leaf tissues infected with Australian, Japanese and Brazilian isolates of Orchid fleck virus (OFV) were indistinguishable and like those previously described in the literature. Cells had an electron-lucent viroplasm with unenveloped rod-shaped virions in the nucleus and cytoplasm, often associated with the inner membrane of the nuclear envelope and the endoplasmic reticulum. Antiserum raised against a Japanese isolate of OFV reacted with Brazilian and Australian isolates in ELISA, … Show more

“…To detect cytopathic effects caused by BTV-C and -N, lesioned tissues were processed, fixed in Karnovsky reagent, embedded in Spurr's epoxy resin and examined by TEM (Kitajima et al, 2001). Leaves were assayed by RT-PCR using specific primers for SvSRV (Ferreira et al, 2007), CoRSV (Kitajima et al, 2011) and ClCSV.…”

“…Plant viruses transmitted by species in the genus Brevipalpus have been described from more than 40 plant species (Kitajima et al 2001(Kitajima et al , 2003Rodrigues et al 2008;. Among these, the citrus leprosis virus complex is by far the most serious (Bastianel et al 2010;Kitajima et al 2011a;Roy et al 2013Roy et al , 2014.…”

Since the taxonomic status for these individual species is uncertain, the identification of which species is/are actually involved in the transmission of CiLV presents a challenge.…”

“…It was first described on Cymbidium sp. in Japan and soon after in several parts of the world in different orchid species and genera (Doi et al, 1977;Freitas-Astúa et al, 1999;Kitajima et al, 2001;Kondo et al, 2003). The viral nature was suggested by the first reported case of N-BTV cytopathology and later by the mechanical infection of some herbaceous species (Doi et al, 1977).…”
