In this study, a detailed chromosome analysis of the endemic Cappadocian Chub, Squalius cappadocicus in Melendiz Stream (Aksaray) was performed. The standard Giemsa staining, C-banding (CBG and CB-DAPI), and Ag-NOR technique were applied. The diploid chromosome number was 50; its karyotype formula was 14M + 16Sm + 10St + 10A. Heteromorphic sex chromosomes weren’t detected in the karyotype of the studied specimens. The number of all chromosomal arms (NF) was 90. In the standard C-banded and CB-DAPI karyotype of the species, dark C-bands were observed in the centromeric region of some bi-armed and acrocentric chromosomes, while slightly centromeric or pericentromeric C-bands were detected in some chromosomes. Three different active Ag-NORs, which were hemizygous, were detected in all samples examined. Two of these active NORs were detected in the bi-armed and the other in the acrocentric chromosome short arm. The Ag-NOR number of this species was evaluated as a feature that distinguishes it from other Squalius species in Türkiye.

Öz Üç tarafı denizlerle çevrili ve yüzlerce tatlı su kaynağı bulunan Türkiye'de tatlı su ve deniz türleri dahil olmak üzere toplam dokuz yüz üç balık türü olduğu bildirilmiştir. Türkiye balık sitogenetiğinde son otuz beş yılda, toplam balık türlerinin yaklaşık %11'ine karşılık gelen, 38'i endemik tür olan ve 17'den fazla takımda yer alan 103 tür/alttürün kromozom özellikleri incelenmiştir. Tüm bu çalışmalarda geleneksel sitogenetik yöntemler kullanılarak (Giemsa, NOR-, C-, GTG-, Q-ve RE boyama-bantlama) balıkların karyotipleri, kromozom sayıları ve yapıları, kol sayıları (NF) ve bazı gen bölgeleri (NOR, heterokromatin vb.) ortaya çıkarılmıştır. Bununla birlikte, Türkiye'de ilk defa üç endemik cyprinid türünün kromozomlarında CMA3 ve DAPI boyama da yapılmıştır. 48 balık türünün kromozomlarında tekrarlayan DNA ve heterokromatik bölgeler NOR ve C-bantlama ile belirlendi. Türkiye'deki diğer balık gruplarına kıyasla en çok görülen Cypriniformes takımının üyesi (Cobitidae, Nemacheilidae, Cyprinidae ve Leuciscidae familyalarından) 69 türün sitotaksonomik ve sitogenetik analizi yapılmıştır. Bu sonuçlara göre sitogenetiksel incelenen balıklarda çoğunlukla kromozom sayısı 2n = 50 olduğu tespit edilmiştir. En yüksek kromozom sayısının 2n = 150 olduğu ve en düşük kromozom sayısının iç sularda doğal olarak bulunan bir Bagridae türüne (2n = 32) ait olduğu da kaydedilmiştir.

The karyotype and distribution of constitutive heterochromatin and nucleolus organizer regions (NORs) of Anatolian leuciscine endemic to Lake Beysehir, Squalius anatolicus (Bogutskaya, 1997) were analyzed respectively using conventional Giemsa-staining, C-banding and Ag-impregnation. Diploid chromosome number was 2n = 50 and karyotype consisted of 7 pairs of metacentric, 13 pairs of submetacentric, 5 pairs of subtelo- to acrocentric chromosomes, NF value equaled 90. Heteromorphic elements indicating sex chromosomes were not detected. C-banding revealed clear pericentromeric constitutive heterochromatin blocks in several chromosomes. Ag-impregnation revealed the size heteromorphism of NORs that covered almost the entire short arms of the middle-sized submetacentric chromosome pair. The karyotype pattern and simple NOR phenotype of S. anatolicus are nearly identical with that found not only in Squalius species analyzed to date but also in many other representatives of the Eurasian leuciscine cyprinids, which indicates remarkable chromosome stasis in this leuciscid lineage.
