Comparative characteristics of the VP7 and VP4 antigenic epitopes of the rotaviruses circulating in Russia (Nizhny Novgorod) and the Rotarix and RotaTeq vaccines

Abstract: Two live, attenuated rotavirus A (RVA) vaccines, Rotarix and RotaTeq, have been successfully introduced into national immunization programs worldwide. The parent strains of both vaccines were obtained more than 30 years ago. Nonetheless, only very limited data are available on the molecular similarity of the vaccine strains and their genetic relationships to the wild-type strains circulating within the territory of Russian Federation. In this study, we have determined the nucleotide sequences of the genes enco… Show more

“…Cytotoxic T lymphocytes (CTL) have been linked to clearance of RV infection resulting in complete short-term and partial long-term protection against re-infection 27 . Two linear CTL epitopes responsible for this activity have been reported on VP7 protein at amino acid positions 16-28 and 40-52 28,29 . An analyses of the Rwandan G1 pre-and post-vaccine introduction strains showed three amino acid differences at positions T41F/S, V42M and A46V with G1 component of the RotaTeq and Y41F/S, V42M and A46V to Rotarix vaccines ( Fig.…”

“…Strains isolated from Russia (RVA/Human-wt/RUS/NN2406/2011/G2P[4] and RVA/Human-wt/RUS/NN2578/2011/G2P[4]) had the same S242N change in the VP7 epitope (7-1b) as RVA/Human-wt/MOZ/0308/2012/G2P[4], which is also similar to RotaTeq ® (SC2-9) G2 [ 39 ]. These strains formed a sub-cluster in lineage IV, separate from other G2P[4] strains (Fig.…”

“…The two established VP7 cytotoxic T-cell lymphocyte epitopes at positions 16-28 and 40-52 [44] of the VP7 of South African G1P [8] strains were compared with cognate regions of Rotarix ® VP7. Five amino acid differences appeared before Rotarix ® introduction, while two amino acid substitutions (A43V in two strains and A46T in four strains) were found after vaccine introduction (Table S1 in S6).…”
