Comparative Benefits and Harms of Second-Generation Antidepressants: Background Paper for the American College of Physicians

Abstract: Current evidence does not warrant the choice of one second-generation antidepressant over another on the basis of differences in efficacy and effectiveness. Other differences with respect to onset of action and adverse events may be relevant for the choice of a medication.

Because of their better tolerability (6), similarity in terms of efficacy and better safety in overdose, second-generation antidepressants including Selective Serotonin Reuptake Inhibitors (SSRIs) have gradually substituted tricyclic antidepressants (TCAs) since mid-1980s and became the first-line medications for the treatment of depression (7).
