Comparative analysis reveals Ce3D as optimal clearing method for in toto imaging of the mouse intestine

Abstract: Background The intestinal wall has a complex topographical architecture. The multi‐layered network of the enteric nervous system and its intercellular interactions are difficult to map using traditional section‐based or whole‐mount histology. With the advent of optical clearing techniques, it has become feasible to visualize intact tissue and organs in 3D. However, as yet, a gap still needs to be filled in that no in‐depth analysis has been performed yet on the potential of different clearing techniques for th… Show more

“…In recent years, a technique called "tissue clearing" has reemerged, offering an alternative approach for tissue sectioning. Nowadays, numerous protocols allow optical clearing and detailed 3D imaging of intact organs [17][18][19][20][21][22].…”

“…iDISCO is an optical clearing method that makes biological samples more transparent ("cleared") ( Fig. 2) and has been successfully used to image three-dimensional structures, including intact mouse organs such as the brain, kidney, intestine, eye, and even whole embryos [18,[22][23][24][25]. Recently, Becher et al [26] applied the iDIS-CO technology for 3D profiling of atherosclerotic plaques and arterial remodeling after carotid artery ligation.…”

“…Immunostaining and clearing procedures for the full-thickness intestinal wall and muscle layer samples were performed as previously described 130 . In brief, whole-mount muscle layer and intestinal wall preparations of LI specimens from psA30P mice and corresponding WT were permeabilized for 4 h at 37°C on a shaker with permeabilization solution (0.01% NaN3 + 1% normal donkey serum + 1% Triton-X-100 in PBS).…”

“…Previously, CytoVista was reported to cause only minor shrinkage. However, these studies referred to the clearing of tissues and not to small 3D structures such as spheroids (Hama et al, 2015;Ke et al, 2016;Bossolani et al, 2019). Also, with Clear T2 , spheroids of human dermal fibroblasts (Costa et al, 2018b) and rodent-derived glioblastoma or neural stem cells (Dingle et al, 2015) showed no significant alterations of sample size.…”
