Comparative analysis of nuclear proteins of B cells in different developmental stages

Abstract: The developmental stage-specific regulation of V(D)J recombination, a gene rearrangement process of antigen receptor gene segments, is tightly controlled in cells. Here we screened proteins uniquely or differentially expressed among three developmentally distinguishable B cells (pro-B, pre-B and mature B cells) using two-dimensional gel electrophoresis and mass spectrometry. Chromatin assembly factor 1 was uniquely expressed in pro-B cells. Purine nucleotide phosphorylase, LCK, E2A and many other unidentified … Show more

“…For protein identification, we used a Voyager -DE™ (delayed extraction) STR biospectrometry workstation (Applied Biosystems, Forster city, CA, U.S.A.) for MALDI-TOF mass spectrometry. The gel pieces containing the desired protein spots were excised, washed and digested in gel with trypsin (sequencing grade, Boehringer Mannheim) (11). The resulting peptides were then processed for MALDI-TOF/MS.…”

“…Proteomic approaches using two-dimensional gel electrophoresis allow one to identify proteins differentially expressed at each B lymphocyte developmental stage. According to the previous study [22], not only some proteins known as B cell lineage-specific factors such as E2A and LCK, but also some other proteins have been newly identified. Effort to isolate and identify proteins differentially expressed in the different stage during the B cell development is undergoing.…”
