Comparative analysis of naive, primed and ground state pluripotency in mouse embryonic stem cells originating from the same genetic background

Abstract: Mouse embryonic stem cells (mESCs) exist in a naive, primed and ground state of pluripotency. While comparative analyses of these pluripotency states have been reported, the mESCs utilized originated from various genetic backgrounds and were derived in different laboratories. mESC derivation in conventional LIF + serum culture conditions is strain dependent, with different genetic backgrounds potentially affecting subsequent stem cell characteristics. In the present study, we performed a comprehensive characte… Show more

“…Compared to ESCs, we found that the majority of pluripotency markers such as Rex1, Klf4, Sox2 and Nanog were downregulated in EPI aggregates, but Oct4 expression was at similar levels. Furthermore, the expression of the epiblastspecific markers Fgf5 and Otx2 was highly upregulated in EPI aggregates, indicating a transition from the naïve to the primed state of pluripotency (Ghimire et al 2018) (Fig. 1d).…”

“…The loss of Dppa3 has no obvious effects on either germ cell determination. 26 Dppa3-positive ES cells have more active histone modification and lower DNA methylation. Dppa3-deficient females failed to produce offspring.…”

“…The detection of these markers is usually performed using flow cytometry, RT-qPCR, and cell-staining techniques 100 . A number of markers can be used to identify PSC types, such as the naive PSC state or the high-yield expansion PSC state [101][102][103][104] , and to measure the quality of the PSCs 105 . At the single-cell level, pluripotency can be assessed with scRNA-seq and bioinformatics tools.…”
