Comparative Analysis of Cellular Prion Detection by Mass‐Sensitive Immunosensors

Abstract: We performed a comparative study of the detection of human cellular prions (PrPC) with a quartz crystal microbalance (QCM) immunosensor using various antibodies against PrPC. The sensing layer was formed by immobilized antibodies on a surface of polyamidoamine dendrimers (PAMAM) of fourth generation (G4) with attached protein A. The sensitivity of detection depended on the kind of antibody used and the best detection limit was 0.8 nM. PrPC detection in 10 times diluted blood plasma revealed more complicated be… Show more

“…The paper by Poturnayova et al 2014, discussed the immobilization of aptamers or monoclonal antibodies for detecting of PrP-C by electrochemical quartz crystal microbalance (EQCM) sensitive to 20 pM for antibodies and 50 pM for DNA aptamers [12]. Small amounts of surfactant tween n phosphorous buffers aided the binding of PrP-C to antibodies protein A (PrA) and antibodies (PRI 308).…”

“…The early diagnostics of PrP Sc is crucial for successful therapy of prion diseases. The determination of the prion diseases is based on immunodiagnostic procedure and by post mortem histopathological identification of brain tissues for example by staining using Congo red and other methods (see and references herein). The attempts to develop of ante mortem assay for detection prion diseases in humans or animals is in a very beginning stage.…”

“…Depending on the type of antibodies one can expect different immunosensor sensitivity. In our recent work we analyzed how the sensitivity of detection of PrP C will depend on the kind of antibody used . For this purpose we used the piezoelectric method allowing also determination of kinetics binding constants.…”
