The present study evaluated the antimutagenic and anticancer activities of A. marina leaf extract. The antimutagenic activity of ethanol and water extracts of A. marina leaf (concentration 10 mg/plate for 48 h) was evaluated using the Salmonella typhimurium TA100 strain in the Ames test with (+S9) and without (−S9) metabolic activation, and its anticancer effect with concentrations of 200, 400, 450, 500, 600, and 800 µg/ml of ethanol extract for 24, 48, and 72 h on human leukaemic cell line HL-60 was assessed by MTT assay. The ethanol and water extracts of A. marina were revealed to have antimutagenic activity in the Salmonella mutation assay. The number of mutant colonies decreased in the presence of ethanol and water extract with (+S9) and without (−S9) metabolic activation. The ethanol extract showed a higher antimutagenic effect than the water extract, with an inhibition rate of 71% on the mutated bacterium. The MTT cell viability test showed an induction of cytotoxicity in HL-60 cells treated with the extract with IC50 values of 600, 400, and 280 µg/ml after 24, 48, and 72 h, respectively, in a concentration and time-dependent manner. Flow cytometric analysis confirmed the induction of apoptosis in the cells. Collectively, these results suggest that the Avicennia marina leaf extract contains bioactive compounds with antimutagenic and antileukaemic effects.

Peptides selected from artificial peptide libraries have special interest in cell targeting, drug discovery, molecular diagnosis and multidrug reversal. Peptides selected from artificial peptide libraries against doxorubicin resistant K562 cells have the capacity in the reversal of the doxorubicin resistance. Our aim is to determine the characteristics of these peptides on recombinant human P-glycoprotein membrane fractions to be able to understand their interactions. Peptides were selected against doxorubicin resistant K562 cells. The effects of synthesized peptides on these cells viability was done by XTT viability assay. The interactions of peptides with P-glycoprotein were done by Pgp-Glo TM Assay System by measuring the ATPase activity of P-glycoprotein. According to our results; four selected peptides effected on doxorubicin resistant K562 cells viability and stimulated the P-glycoprotein activity in the presence of doxorubicin at different levels. If the P-glycoprotein stimulation occurs on living doxorubicin resistant K562 cells, cell viability will not be affected due to multidrug resistance effect of P-glycoprotein. Peptides selected from artificial peptide libraries are useful tools in the reversal of multidrug resistance and for understanding the membrane structure-function relationships.

Problem statement: Leukemia is a cancer which linked intimately with the immune system. It is a group of heterogenous neoplastic disorder of white blood cells characterized by the uncontrolled proliferation and block in differentiation of hematopoeitic cells. Generally, leukemia can be classified into myeloid or lymphoid referring to the type of abnormal cell found most in the blood. Approach: Many drug discovery researches have utilized the leukemic cell lines to screen for their potential as chemotherapy agents for leukemia. Results: The understanding on the types of leukemia and current choice of chemotherapy drugs can help to select the best target cell lines to study and the most suitable chemotherapy drug as positive control. Conclusion/Recommendations: This review cover types of leukemia, current option of chemotherapy drugs and some potential cytotoxic and immunomodulatory natural products targeting leukemic cancer cell line.
