Comparative Analysis of African and Asian Lineage-Derived Zika Virus Strains Reveals Differences in Activation of and Sensitivity to Antiviral Innate Immunity

Abstract: In recent years, Asian lineage Zika virus (ZIKV) strains emerged to cause pandemic outbreaks associated with a high rate of congenital ZIKV syndrome (CZVS). The reasons for the enhanced spread and severe disease caused by newly emerging strains are not fully understood. Here we compared viral sequences, viral replication, and innate immune signaling induction of three different ZIKV strains derived from African and Asian lineages and West Nile virus, another flavivirus. We found pronounced differences in activ… Show more

“…To address this question, we infected our microglia-containing organoids with two different stains of ZIKV, MR766 and Fortaleza. Consistent with previous studies, ZIKV-MR776 exhibited greater virulence than Fortaleza (Esser-Nobis et al, 2019), as indicated by the significant morphological abnormalities and cell detachment in the organoid ( Figure 6A, S3B). Thus, we chose to use the MR766 strain for the following experiments.…”

“…ZIKV is classified into two major lineages or strains, the Asian/American and African. Several studies have shown various differences between the two strains, such as differences in infection rate, viral particle production, cellular and antiviral responses and sensitivity to antiviral immunity [30,31]. Differences in pathogenicity has also been observed between the various subtypes of the Asian/American lineage [32].…”

“…Further, these responses vary among the different ZIKV strains isolated from different geographical locations. ZIKV strains from Brazil and Uganda showed delayed activation of the innate immune responses mediated by RIG-I as compared to the milder Cambodia strain that correlates with their pathological outcomes ( Esser-Nobis et al, 2019 ). Studies using these different strains in lung A549 cells revealed the important role of RIG-I sensing in early innate immune response and induction of Type I Interferon responses ( Strottmann et al, 2019 ).…”
