Comparable dose-dependent inhibition of AP-7 sensitive strychnine-induced allodynia and paw pinch-induced nociception by mexiletine in the rat

Abstract: The blockade of spinal glycine receptors with intrathecal (i.t.) strychnine produces segmentally-localized allodynia in the rat; a reversible and highly reproducible effect that is attained without peripheral or central nerve injury. We investigated the effect of i.v. mexiletine, an orally active congener of lidocaine, on strychnine allodynia and compared the dose-response relationship of mexiletine in normal (noxious paw pinch) versus abnormal (i.t. strychnine) nociceptive conditions. In addition, we determin… Show more

“…GABAergic transmission to lamina II neurons was reduced after two distinct partial peripheral nerve lesions, CCI and SNI, both of which generate marked pain hypersensitivity. Pharmacological antagonism of GABAergic and glycinergic inhibitory transmission in the spinal cord in normal animals also generates a neuropathic pain-like tactile hypersensitivity (Yaksh, 1989;Sivilotti and Woolf, 1994;Khandwala et al, 1997). These two observations raise the key question: does diminished inhibitory transmission in the dorsal horn contribute to heightened pain sensitivity after partial nerve injury?…”

Notably, the 5-HT 3 receptor belongs to the same super family of ligand-gated ion channels as nicotinic acetylcholine, GABA A and glycine receptors.…”

“…A mexitelina é utilizada VO e exerce atividade anestésica, antiarrítmica e analgésica em dores lancinantes de origem neuropática (neuralgia do trigêmeo, outras neuralgias da face, neuralgia pós-herpética, neuralgia diabética) 95 . Apresenta propriedades farmacológicas semelhantes às da quinidina e da procainamida.…”
