Common Lung Microbiome Identified among Mechanically Ventilated Surgical Patients

Abstract: The examination of the pulmonary microbiome in patients with non-chronic disease states has not been extensively examined. Traditional culture based screening methods are often unable to identify bacteria from bronchoalveolar lavage samples. The advancement of next-generation sequencing technologies allows for a culture-independent molecular based analysis to determine the microbial composition in the lung of this patient population. For this study, the Ion Torrent PGM system was used to assess the microbial c… Show more

“…The bacterium Ruegeria pomeroyi is more abundant in corals with white patch syndrome or skeletal growth anomalies than in healthy corals (Sere et al, 2013 ; Li et al, 2015 ). The genus Thauera , of the class Betaproteobacteria , has been detected in sediments, sludge, soils, and wastewater treatment plants (Liu et al, 2013 ), and it has been recently reported in research on human (Smith et al, 2016 ; Xu et al, 2017 ). Xu et al ( 2017 ) observed that Thauera was significantly more abundant in individuals with substance use disorders than in healthy individuals.…”

“…Characteristics of each sample group regarding age, gender, prior antibiotic administration, reason for BAL, and days post injury for BAL collection are listed in Table 1 . Sequence analysis of 6 of the C- samples were previously reported in Smith et al [ 24 ]. Samples were selected sequentially based upon the diagnosis of the pathology laboratory.…”

“…As described previously [ 24 ], upon collection, a portion of the raw BAL fluid was retained for research analysis and stored in a polypropylene tube at 4°C. The remaining BAL fluid was submitted to the Parkland Memorial Hospital Clinical Pathology Laboratory for microbiological identification as part of the standard of care.…”

“…Trying to identify the best methodology, we believed it was crucial to use different comparators or positive controls: an internal bacterial positive control (mock population) and conventional microbiology. Because many studies did not do so, it seems hard to interpret the results of many studies revealing bacterial genus or species which are usually not identi ed in respiratory samples [32] [33]. It is thereby impossible to determine which methodology or primer set give the most reliable results.…”
