Coming together to define membrane contact sites

Abstract: Close proximities between organelles have been described for decades. However, only recently a specific field dealing with organelle communication at membrane contact sites has gained wide acceptance, attracting scientists from multiple areas of cell biology. The diversity of approaches warrants a unified vocabulary for the field. Such definitions would facilitate laying the foundations of this field, streamlining communication and resolving semantic controversies. This opinion, written by a panel of experts i… Show more

“…Using electron microscopy 46 , we found no difference in the percentage of endosomes in close proximity (>30 nm) to ER between wild type and TMEM16K knockout cells (Supplemental Fig. 5a,b), consistent with the presence of multiple proteins maintaining these contacts 47,48 . We have further strengthened these observations with proximity ligation assay (PLA) in situ, a powerful novel approach to study contact sites alterations in a quantitative manner.…”

“…The identification of structures allowing membrane tethering of two organelles into close proximity has instigated considerable research interest into the (patho)physiologic role of inter-organelle contact sites (reviewed in [79]). Indeed, the juxtaposition (<30nm) of membranes of identical (homotypic contacts) or distinct organelles/membrane types (heterotypic contacts) acts as a domain for inter-organelle communication [79].…”

“…A distance threshold ≤ 30 nm was chosen as representative of an interaction between nearby organelles, on the basis of previously established morphometric analysis in animal and plant cells (Helle et al, 2013;Mueller-Schuessele and Michaud, 2018;Scorrano et al, 2019). Red: original image.…”
