Combined treatment with ultrasound-targeted microbubble destruction technique and NM-aFGF-loaded PEG-nanoliposomes protects against diabetic cardiomyopathy-induced oxidative stress by activating the AKT/GSK-3β1/Nrf-2 pathway

Abstract: The present study sought to investigate the effect of non-mitogenic acidic fibroblast growth factor (NM-aFGF) loaded PEGylated nanoliposomes (NM-aFGF-PEG-lips) combined with the ultrasound-targeted microbubble destruction (UTMD) technique on modulating diabetic cardiomyopathy (DCM)and the mechanism involved. Animal studies showed that the diabetes mellitus (DM) group exhibited typical myocardial structural and functional changes of DCM. The indexes from the transthoracic echocardiography showed that the left v… Show more

“…For instance, liposomes loaded with a non-mitogenic acid fibroblast growth factor and associated to a microbubble are designed to be precisely destroyed with the targeted ultrasound. This approach led to an improved systolic function and milder structural remodeling in STZ-treated rats ( 135 ). Effective cardiac gene delivery using the natural tropism of adeno-associated virus serotypes 9 and 6 toward the DCM myocardium are described ( 89 , 136 , 137 ).…”

“…Cardiomyopathy, which is characterized by the presence of a dysfunctional heart muscle preventing the heart from properly pumping blood to other parts of the body, is described as occurring in patients suffering from diabetes or treated with certain drugs such as doxorubicin, [11,26,29,36,37,46,111]. Combining nanometric contrast agents (MBs) with a targeting ligand (FGP1) makes it possible to bring FGP1 specifically in the DHR, where FGP1 can promote endothelial vascular tissue/smooth muscle cell proliferation.…”

“…(DSPE-PEG2000) were dissolved in a mixture of chloroform and methanol in a molar ratio of 4:5:1, and then was operated by a JY 92-II ultrasonic equipment at 30°C for 3 min (frequency and power parameters:40 kHz, 160 W). After the cationic lipid microbubbles solution was frozen at −20°C in the refrigerator and was lyophilized in vacuum freeze dryer at pressure of 15 Pa (at −48°C for 15 h, and then up to 10°C for no more than 5 h), the cationic lipid microbubbles (150 mg) were added to the mixture of 0.9% normal saline and glycerol (volume ratio 4:1.5 ml) for dispersion (Zhang et al, 2020). The liquid was transferred to a borosilicate glass bottle and filled with inert gas SF6.…”
