Significance of Light, Sugar, and Amino Acid Supply for Diurnal Gene Regulation in Developing Barley Caryopses

The caryopses of barley (Hordeum vulgare), as of all cereals, are complex sink organs optimized for starch accumulation and embryo development. While their early to late development has been studied in great detail, processes underlying the caryopses' diurnal adaptation to changes in light, temperature, and the fluctuations in phloem-supplied carbon and nitrogen have remained unknown. In an attempt to identify diurnally affected processes in developing caryopses at the early maturation phase, we monitored global changes of both gene expression and metabolite levels. We applied the 22 K Barley1 GeneChip microarray and identified 2,091 differentially expressed (DE) genes that were assigned to six major diurnal expression clusters. Principal component analysis and other global analyses demonstrated that the variability within the data set relates to genes involved in circadian regulation, storage compound accumulation, embryo development, response to abiotic stress, and photosynthesis. The correlation of amino acid and sugar profiles with expression trajectories led to the identification of several hundred potentially metabolite-regulated DE genes. A comparative analysis of our data set and publicly available microarray data disclosed suborgan-specific expression of almost all diurnal DE genes, with more than 350 genes specifically expressed in the pericarp, endosperm, or embryo tissues. Our data reveal a tight linkage between day/night cycles, changes in light, and the supply of carbon and nitrogen. We present a model that suggests several phases of diurnal gene expression in developing barley caryopses, summarized as starvation and priming, energy collection and carbon fixation, light protection and chaperone activity, storage and growth, and embryo development.

Kalanchoë daigremontiana reproduces asexually by generating numerous plantlets on its leaf margins. The formation of plantlets requires the somatic initiation of organogenic and embryogenic developmental programs in the leaves. However, unlike normal embryogenesis in seeds, leaf somatic embryogenesis bypasses seed dormancy to form viable plantlets. In Arabidopsis (Arabidopsis thaliana), seed dormancy and embryogenesis are initiated by the transcription factor LEAFY COTYLEDON1 (LEC1). The K. daigremontiana ortholog of LEC1 is expressed during leaf somatic embryo development. However, KdLEC1 encodes for a LEC1-type protein that has a unique B domain, with 11 unique amino acids and a premature stop codon. Moreover, the truncated KdLEC1 protein is not functional in Arabidopsis. Here, we show that K. daigremontiana transgenic plants expressing a functional, chimeric KdLEC1 gene under the control of Arabidopsis LEC1 promoter caused several developmental defects to leaf somatic embryos, including seed dormancy characteristics. The dormant plantlets also behaved as typical dormant seeds. Transgenic plantlets accumulated oil bodies and responded to the abscisic acid biosynthesis inhibitor fluridone, which broke somatic-embryo dormancy and promoted their normal development. Our results indicate that having a mutated form of LEC1 gene in K. daigremontiana is essential to bypass dormancy in the leaf embryos and generate viable plantlets, suggesting that the loss of a functional LEC1 promotes viviparous leaf somatic embryos and thus enhances vegetative propagation in K. daigremontiana. Mutations resulting in truncated LEC1 proteins may have been of a selective advantage in creating somatic propagules, because such mutations occurred independently in several Kalanchoë species, which form plantlets constitutively.

The Dof proteins belong to a large family of plant transcription factors that share a single highly conserved zinc fi nger and play an important role in many physiological processes.To elucidate the function of Dof in soybean, GmDof17-1 was cloned from soybean (Glycine max (L.) Merr); the open reading frame (ORF) of GmDof17-1 is 846 bp and encodes a putative protein that includes 281 amino acids. Using qRT-PCR, the expression profi les of GmDof17-1 were obtained for various parts of the soybean plant. GmDof17-1 was primarily expressed in the roots and pods at various stages of pod development. The gene was ectopically expressed in tobacco under the control of the 2 × CaMV35S promoter to study the functions of the gene product.The transgenic tobacco plants showed to be dwarf, and the indole-3-acetic acid (IAA) content was decreased, whereas the sulfur-containing amino acid content of the seeds increased. These results provide new insight into the function of GmDof17-1 in seed development. IntroductionDof proteins are a major family of plant transcription factors that have a strongly conserved DNA-binding domain, which contains a single C 2 C 2 -type zinc-fi ngerlike motif that is generally located in the N-terminal region and a C-terminal domain for transcriptional regulation (this region has divergent amino acid sequences) (Yanagisawa and Izui, 1993;Yanagisawa, 2004). Since Dof1 (the fi rst identifi ed Dof protein) was found in maize (Yanagisawa and Izui, 1993), many Dof proteins have been discovered, which participate in specifi c biological processes in other plants. The functions of reported Dof genes are highly diverse and include roles in hormone response, defense response, organ development and other responses (Baumann et al., 1999;Kang and Singh, 2001; Kang et al., 2000;Kisu et al., 1998;Mena et al., 1998;Plesch et al., 2001;Vicente-Carbajosa et al., 1997;Wang et al., 2007;Ward et al., 2005;Washio, 2001;Xin et al., 2009;Yang et al., 2010).Phytohormones play important roles in coordinating many growth-related processes throughout the plant life cycle, including embryogenesis, vascular differentiation, organogenesis, tropic growth and root and shoot architecture (Kepinski, 2006;Quint and Gray, 2006; Shan et al., 2012 Stamm andKumar, 2010).
