Combined Effects of Tumor Promoters and Serum on Proliferin mRNA Induction: A Biomarker Sensitive to Saccharin, 2,3,7,8-Tcdd, and Other Compounds at Minimal Concentrations Promoting C3H/10T1/2 Cell Transformation

Abstract: Increases in proliferin (PLF) gene family mRNA abundance and promotional effects in cell transformation assays are paired responses that follow exposures to diverse chemical and physical agents in the C3H/10T1/2 in vitro model of multi-stage carcinogenesis. This study measured PLF mRNA abundance changes over 1 to 3 d in response to several types of promoters that were previously unassessed for this effect. Saccharin is a known promoter of cell transformation in C3H/10T1/2 cell cultures, but unlike 12-O-tetrade… Show more

“…It is possible that the cocarcinogenic properties of formaldehyde and acetaldehyde derive from their toxicity and subsequent stimulation of cell proliferation, , which would increase the initiating potential of NNK. Both acetaldehyde and formaldehyde promote cell transformation in C3 H /10T1/2 cell cultures . These aldehydes also alter gene expression in ways that could enhance tumor initiation by NNK.…”

“…Other genes, such as metallothionin and prostaglandin H synthase‐2, respond to TPA and several other tumor promoters, but not to BPO [43,44]. Mrp/plf ‐mRNA has been shown to increase in vitro in response to 17 different promoters of cell transformation [7] and may therefore parallel the response‐pattern of the more broadly responsive gene group. Additional studies in skin with all of these genes and a broader range of skin tumor promoters will be required to confirm this conjecture.…”

“…The 596 bp cDNA fragment contains only the upstream primer‐site targeted by the mrp/plf ‐specific PCR primer‐pair used in other parts of this study (see below), thereby minimizing chances for spurious amplifications because of cross‐contamination of laboratory ware or PCR solutions. Blot prehybridization, hybridization, washing, and probe stripping procedures were carried out as described previously [7]. Developed autoradiographic images within the linear response range of Kodak BioMax film (optical density 0.2–2.0) were quantified with a Molecular Dynamics 300B scanning/computing densitometer and Image Quant software.…”
