Combined effects of bucillamine and etanercept on a rat type II collagen-induced arthritis model

Abstract: The combined effects of bucillamine (Buc) and etanercept (ETN) on a rat model of type II collagen (CII)-induced arthritis (CIA) after treatment onset were investigated. In the combination treatment, rats received Buc 30 mg/kg orally administered once daily from the onset of arthritis or from 4 days after the onset of arthritis and ETN 0.3 mg/kg subcutaneously administered once on the day of onset. The effects of monotherapy with Buc and ETN, respectively, and of Buc + ETN combination therapy on the resulting p… Show more

“…We initiated treat ment on day 35, a time at which most mice had developed arthritis. This is in contrast to the majority of studies in which the drug being stud ied is administered at or before the initiation of the disease [25][26][27][28][29][30]. If one uses TNFa blockade as an example, treatment at the time of disease initiation results in a profound block in disease.…”

“…DMARDs tested were leflunomide (one experiment),32 methotrexate (four experiments),33–35 anti-tumour necrosis factor (TNF) (two experiments)36 and anti-interleukin (IL)1 (two experiments) 37. A meta-analysis of these studies demonstrated that prearthritis intervention is associated with a reduction of arthritis severity in animal models of arthritis (p=0.005).…”

“…To further characterize our model, we profiled a panel of therapeutic agents such as, etanercept, dexamethasone and buprenorphine. We selected these agents based on their established efficacy in other preclinical models of arthritis [ 55 57 ]. The specific doses used for each drug in these studies were selected based on multiple dose response studies conducted internally (data not shown).…”
