Combined effect of bisphosphonate and recombinant human bone morphogenetic protein 2 on bone healing of rat calvarial defects

Abstract: BackgroundThis study aimed to investigate new bone formation using recombinant human bone morphogenetic protein 2 (rhBMP-2) and locally applied bisphosphonate in rat calvarial defects.MethodsThirty-six rats were studied. Two circular 5 mm diameter bony defect were formed in the calvaria using a trephine bur. The bony defect were grafted with Bio-Oss® only (group 1, n = 9), Bio-Oss® wetted with rhBMP-2 (group 2, n = 9), Bio-Oss® wetted with rhBMP-2 and 1 mM alendronate (group 3, n = 9) and Bio-Oss® wetted with … Show more

“…Group XENO_Low BP ( n = 8) were grafted with xenogenic bone (CollaOss; Bioland, Korea) locally administered in low‐concentration alendronate (1 mM; Cayman Chemical, USA), whereas group XENO_High BP ( n = 8) were grafted with xenogenic bone locally administered in high‐concentration alendronate (10 mM). The concentrations of alendronate used in this study were based on a previous study (Kim, Song, et al., ).…”

“…In previous studies, the concentration used to prevent root resorption in delayed replantation after extraction of teeth was 1 mM (0.325 mg/ml) (Komatsu et al, 2008;Levin, Bryson, Caplan, & Trope, 2001;Shibata et al, 2004). Moreover, another previous study (Kim, Song, et al, 2015) used 1 and 10 mM alendronate as low and high concentration, respectively. Thus, in this study, we used two concentrations of alendronate, 1 and 10 mM.…”

“…Collectively, our data suggest that DC‐STAMP, together with MMP‐2 and MMP‐9 (Galliera et al, ; Rocha et al, ), can be a useful tool for monitoring, in parallel with other bone turnover markers, to evaluate bone remodeling and tissue healing. Given that DC‐STAMP+ cells are homing to bone fracture sites, a step forward approach is to molecular engineer a chimeric DC‐STAMP protein by which DC‐STAMP is able to bring small peptides/proteins such as bone morphogenetic protein 2 (BMP‐2) or MMP‐9 (Kim et al, ; Matsumoto et al, ; Miller et al, ; Saito et al, ; Wang et al, ) with healing‐promoting effects to bone fracture sites.…”
