Combined approach of NMR and chemometrics for screening peptones used in the cell culture medium for the production of a recombinant therapeutic protein

Abstract: Soy peptones or soy hydrolysates are widely used as key medium additives in serum-free cell culture processes for industrial production of therapeutic recombinant proteins. The heterogeneous nature of these vegetable-derived materials can lead to substantial lot- to-lot variability in cell culture processes. In this study, we demonstrated the feasibility of nuclear magnetic resonance (NMR) spectroscopy in combination with chemometrics in rapid screening peptone lots in order to optimize efficiency and consiste… Show more

“…resonance (NMR), 3,4 Raman 5 or two-dimensional (2D)-fluorescence spectroscopy 6 have been proposed to characterize complex raw materials. Those methods generate very large data sets that can only be handled by chemometric methods like principal component analysis (PCA) or partial least squares modeling (PLS).…”

“…Their proposed method demonstrated satisfactory performance with respect to linearity and precision and was able to resolve components with specificity. More recently, Luo and Chen examined use of an approach using a combination of nuclear magnetic resonance (NMR) and chemometrics for screening peptones used in cell culture media (Luo and Chen, 2007). Their approach allowed or rapid screening of peptone lots in order to optimize efficiency and consistency of large-scale protein production.…”

“…However, there are a few limitations that complicate hydrolysates' utilization. First of all, lot-to-lot variation exists for hydrolysates (Luo and Chen, 2007;Zhang et al, 2003). Prescreening using smallscale performance testing is typically required to eliminate underperforming lots.…”
