Combination of treadmill exercise with bone marrow stromal cells transplantation activates protein synthesis-related molecules in soleus muscle of the spinal cord injured rats

Abstract: The present study investigated whether treadmill exercise with bone marrow stromal cells (BMSCs) transplantation increase expression level of protein synthesis-related molecules in the soleus muscle after spinal cord injury (SCI). The spinal cord contusion injury was performed at the T9–10 level using the impactor (10 g×25 mm). BMSCs were cultured from femur and tibia of 4-week-old rats and then transplanted directly into the lesion 1-week post injury. The rats in exercise group were walking on treadmill devic… Show more

“…Previous clinical studies have reported that rehabilitative training increases physical activity levels and enhances the quality of life in people with SCI [ 7 11 ]. The mechanisms contributing to recovery have been partially demonstrated by facilitating synaptic plasticity [ 12 , 13 ], promoting the expression of neurotrophins such as brain-derived neurotrophic factor (BDNF) and insulin-like growth factor 1 (IGF-1) [ 14 17 ], and then activating the downstream signaling pathway such as mTOR [ 14 , 18 , 19 ]. However, previous studies suggest that rehabilitative training alone have cannot promote regrowth of the CST axons [ 20 ].…”

“…SCI model was induced according to the previously described method [12]. The rats were anesthetized by inhalation of isoflurane (2% isoflurane in 30% O2 and 70% N2, JW Pharmaceutical, Seoul, Korea) during surgery.…”

“…Exercise promotes skeletal muscle health in many ways. It affects multiple physiological processes involved in molecular, cellular, and biochemical pathways regulating skeletal muscle gene expression and the metabolic process, thereby promoting the health of skeletal muscle (11,20,27). Sarcomeres are represented as the elemental contractile units of striated muscles and are composed of thick filaments, thin filaments, and titin filaments.…”
